    Abstract: A sample purification system includes a container assembly bounding a sample purification compartment and having an upper end and an opposing lower end, the sample purification compartment comprising mixing zones and settling zones. A plurality of shielding elements are positioned within the sample purification compartment so as to at least partially separate adjacent mixing zones and settling zones or separate adjacent mixing zones, the mixing zones being in fluid communication with the settling zones. A mixing element is disposed within each mixing zone. An acoustic wave settler is aligned with a portion of the container assembly, the acoustic wave settler being configured to emit an acoustic wave through the portion of the container assembly and a mixture disposed therein, the acoustic wave coalescing fluid phase droplets disposed in the mixture to increase the buoyancy or density of the fluid phase droplets.

    Abstract: A targeted panel with low sample input requirements from a tumor only sample may be processed to estimate mutation load in a tumor sample. The method may include detecting variants in nucleic acid sequence reads corresponding to targeted locations in the tumor sample genome; annotating detected variants with an annotation information from a population database; filtering the detected variants, wherein the filtering rule set retains the somatic variants and removes germ-line variants; counting the identified somatic variants to give a number of somatic variants; determining a number of bases in covered regions of the targeted locations in the tumor sample genome; and calculating a number of somatic variants per megabase, provides an estimate of the mutation load per megabase in the tumor sample genome.

    Abstract: In one aspect, a system for implementing a copy number variation analysis method, is disclosed. The system can include a nucleic acid sequencer and a computing device in communications with the nucleic acid sequencer. The nucleic acid sequencer can be configured to interrogate a sample to produce a nucleic acid sequence data file containing a plurality of nucleic acid sequence reads. In various embodiments, the computing device can be a workstation, mainframe computer, personal computer, mobile device, etc. The computing device can comprise a sequencing mapping engine, a coverage normalization engine, a segmentation engine and a copy number variation identification engine.

    Abstract: A thermal cycler system for use with a sample holder configured to receive a plurality of samples includes a sample block configured to receive the sample holder, a cover lid configured to move in a direction toward the sample block from an open position to a closed position, a heated cover operatively coupled to the cover lid and configured to move in a direction toward the sample block from a raised position to a first lowered position, in which the heated cover contacts the sample holder when the sample holder is received by the sample block, and a drive assembly including a motion guide configured to move in a direction toward the sample block from a first position, wherein the cover lid is in the open position and the heated cover is in the raised position, to a second position, wherein the cover lid is in the closed position.
