An external fixation assembly includes a plurality of hollow pins that are inserted into a patient's bone. Each pin has an interior bore and a plurality of apertures extending through the pin wall from the bore. The pin may be coupled to a source of vacuum pressure operable to create reduced pressure in the tissue surrounding the pin. A cover is placed around the pin and sealed to provide a fluid-tight enclosure that maintains reduced pressure around the pin. A method for applying external fixation using the fixator pins described above includes the steps of inserting each pin through a skin opening, positioning the pin apertures near selected tissue, covering the skin opening with a sealed enclosure, connecting the pins to a source of vacuum pressure, and activating the source of vacuum pressure to create reduced pressure in the patient's tissue at or near the bone.

A topical wound treatment composition comprises a hydrogen peroxide generator; alkaline powder; not more than 5 percent by weight of water; additional topical active agent if desired, and emollient (preferably hygroscopic emollient) to balance. When topically applied to a wound and water from the surrounding environment diffuses into the composition, the hydrogen peroxide generator and/or the alkaline compound diffuse into one another, causing a chemical reaction that generates treatment-effective amounts of oxygen to occur. The oxygen can then diffuse out of the composition and aid in wound treatment or healing.

The present invention provides methods of identifying a subject as responsive to treatment of an inflammatory disease and/or inflammation with one or more polyunsaturated fatty acids (PUFAs), comprising detecting, in a nucleic acid sample of a subject having an inflammatory disease and/or inflammation, the presence of a C allele at single nucleotide polymorphism (SNP) rs730012 in the leukotriene C4 synthase gene, wherein the presence of said C allele at rs730012 in said nucleic acid sample from the subject indicates that the subject is responsive to treatment of said inflammatory disease and/or inflammation with one or more PUFAs.

MEX3C deficiency impairs the development of white and brown adipose tissue. Hence the present invention provides, among other things, a method of screening a candidate compound for activity in inhibiting fat deposition in a subject in need thereof and/or treating a condition in a subject in need thereof, comprising: (a) contacting a candidate compound to a cell that expresses MEX3C protein; and then (b) detecting a quantity of expression of the MEX3C protein in the cell; a depression in the expression of MEX3C protein when the candidate compound is contacted thereto as compared to that expressed when the candidate compound is not contacted thereto indicating the compound is active in inhibiting fat deposition and/or treating a condition in a subject in need thereof. Methods of treatment and screening subjects are also described.

In certain embodiments, the invention provides compositions and methods for treating, improving muscle function, and prolonging survival in a subject with X-linked myotubular myopathy (XLMTM). The present invention provides a method comprising systemic administration of a composition that induces the increased expression of myotubularin in the muscle of a subject. The invention provides sustained regional and global increases in muscle function.
