The invention provides enantiomers of ajoene and derivatives of Z(−)-ajoene. The derivative of Z(−)-ajoene are useful for modulating integrin-mediated functions, for treating disorders, diseases or conditions in which integrins play a role, and for treating tissues to improve their condition for a subsequent use, such as transplantation.

Ratcheting tools are described that include: (a) a tool body; (b) a driven element; (c) a ratchet mechanism coupling the driven element to the tool body, wherein the ratchet mechanism is configured for adjustment between a plurality of a forward configuration, a reverse configuration, and a neutral configuration; (d) an adjustable first biasing element coupled to the ratchet mechanism and configured to bias the ratchet mechanism to at least one of the forward, reverse, and neutral configurations; and (e) an adjustable second biasing element coupled to the ratchet mechanism and configured to bias the ratchet mechanism to at least one of the forward, reverse, and neutral configurations.

A nutritional supplement composed of phytosterols, anti-oxidants, and other complexes, including hydrolysates of fatty acids, amino acids, peptides, proline rich polypeptides and digestive enzymes is described. The nutritional supplement may be used by individuals suffering from or at risk of developing immune system diseases; breast cancer, colon and prostate cancer; HIV infection; or enlarged prostate. The present invention relates generally to systems and methods for assessing blood flow in blood vessels, for assessing vascular health, for conducting clinical trials, for screening therapeutic interventions for adverse effects, and for assessing the effects of risk factors, therapies and substances, including therapeutic substances, on blood vessels, especially cerebral blood vessels, all achieved by measuring various parameters of blood flow in one or more vessels and analyzing the results in a defined manner. The relevant parameters of blood flow include mean flow velocity, systolic acceleration, and pulsatility index. By measuring and analyzing these parameters, one can ascertain the vascular health of a particular vessel, multiple vessels and an individual. Such measurements can also determine whether a substance has an effect, either deleterious or advantageous, on vascular health. The present invention further provides an expert system for achieving the above.

In some embodiments, the disclosure relates generally to methods as well as related compositions, systems, kits and apparatus comprising linking proteins to target compounds and/or to locations of interest using tethers. For example, the tether can be used to link the protein to a target compound, for example, to link an enzyme to a substrate. Similarly, the tether can be used to link the protein at or near a desired location on a surface. In one group of embodiments, the tether includes a polynucleotide and the target compound or location on the surface includes another polynucleotide that is capable of hybridizing to the tether. In such embodiments, the tether can be used to link the protein to the target compound or location using nucleic acid hybridization.

A tool of the type having a drive stud for receiving and releasing a tool attachment includes an opening in the drive stud and a locking pin movably mounted in the opening. The opening defines first and second ends, and the first end of the opening is located at a portion of the drive stud constructed for insertion into the tool attachment. An actuating member is movably positioned on the drive stud, and the actuating member defines a sliding surface that engages the pin. A first spring biases the sliding surface toward the pin, and a second, weaker spring biases the pin toward the sliding surface. The first spring reacts against a ring that is disposed between the spring and a shoulder formed on the tool. The shoulder extends away from the longitudinal axis of the drive stud by a lesser distance than the spring or the ring.

Methods, compositions, systems, apparatuses and kits comprising modified proteins, particularly modified nucleic acid-binding proteins with altered buffering properties are provided. For example, in some embodiments, methods of forming modified proteins including one or more amino acid modifications to achieve desired pKa values are described. Furthermore, the invention provides methods for using such modified proteins in ion-producing reactions, such as ion-based nucleic acid sequencing reactions.
