A thermally stable tryptophanase having (1) an optimum temperature for activity at a pH of 8.0 of about 70.degree. C., and (2) such thermal stability that it is not thermally deactivated when maintained at temperatures up to about 65.degree. C. and a pH of 8.0 for 40 minutes. The thermally stable tryptophanase can be produced by cultivating in a tryptophan-containing culture medium a thermally stable tryptophanase-producing bacterium which does not grow alone in said medium but grows there in the presence of Bacillus sp. strain S, and obtaining the resulting thermally stable tryptophanase from the culture broth. The thermally stable tryptophanase-producing microorganism for use in the above process is a novel organism.

L-tryptophan can be prepared in good yield by a fermentation process which comprises culturing a novel L-tryptophan-producing microorganism of the genus Corynebacterium, which is resistant to at least one member selected from glyphosate [N-phosphonomethyl glycine], paraquat [1,1'-dimethyl-4,4'-bispyridinium] and derivatives thereof, and recovering L-tryptophan from the culture broth.

Biochemical optical resolution of DL-tryptophanes in which DL-tryptophane amides are interacted with the culture products, or their treated products, of a microorganism capable of producing amidase is described. L-tryptophane amides in racemic DL-tryptophane amides are asymmetrically hydrolyzed to form optically active L-tryptophanes at a high yield and almost all D-tryptophane amides remain without being subjected to hydrolysis. The resultant D-tryptophane amides are readily hydrolyzed, after separating L-tryptophanes, to form optically active D-tryptophanes at a high yield.
