摘要: To detect bands in an electrophoresis capillary tube having a liquid separating medium, a light source and a light detector of a monitor are positioned on opposite sides of the capillary tube with the light source transmitting light through a first slit and light passing to the detector through a second slit after it has passed through the capillary tube. The first and second slits are aligned with the direction of motion of bands, have a maximum length of less than 500 micrometers and a maximum width of less than 200 micrometers. There are no vertical lengths in the capillary tube having a dimension greater than one third of its length between a sample injecting end and a detecting end of the capillary tube. A sample injector includes a vacuum chamber communicating with one end of the capillary tube to pull sample into the other end and a pressure sensor to measure the pressure in said vacuum chamber and generate a signal indicative thereof and integrate it to indicate the amount of sample pulled into the system.

摘要: To separate high-molecular DNA strands, an electrophoresis system includes first means for changing the direction of an electric field within an electrophoresis separating unit at a first frequency having a period of no more than 20 seconds and second means for changing the electric field in another manner at a second frequency at least twice as high as the said first frequency. In one embodiment, the field strength in the direction of overall migration is selected so that, considering the DNA size and gel density, the leading segment of DNA to the first corner is sufficiently small so that excess friction forces due to bunching does not cause excessive band spreading. The field strength is selected so that the electric field is reduced until the changing of the electric field at the second frequency keeps the bunching effect to a sufficiently low level to not seriously interfere with the separation.

摘要: An apparatus for supercritical fluid extraction incorporates a removable extraction cartridge which in operation has insignificant pressure difference between its inside and outside walls. In one embodiment, the extractor includes a fraction collector for extractants, an automatic sample changer and an automatic cartridge transfer mechanism which provide completely automated extractions. To automatically perform extraction, valves for the fluids are automatically opened and closed in synchronism with the insertion and removal of the cartridges. These valves force a hard valve element into a softer valve seat with a valve stem that does not rotate significantly under the control of a rotary motor.

摘要: To avoid deposits on the restrictor that channels extractant into a collector, a heated capillary tube pressure release restrictor, has a thermally insulated outlet end in a collecting trap substantially colder than the capillary tube. The restrictor is heated between the insulation and the capillary tube by Joulean heating. The solvent in the trap is at a pressure of 5 to 200 psi above atmospheric pressure. The thermal resistance of the insulation is selected to reduce the heat added to the extractant to a minimum and yet cause the temperature of the extractant to be in a range within which it is sufficiently hot so it does not freeze when added to the collection solvent but not so hot as to reduce partitioning of the extract and extractant before the extractant leaves the collection solvent. It has a thermal conductivity no greater than 60 BTU's per hour, per square foot, per inch for a one degree Fahrenheit difference.

摘要: An apparatus for supercritical fluid extraction incorporates a removable extraction cartridge which in operation has insignificant pressure difference between its inside and outside walls. Because of the low pressure difference, the extraction cartridge need not have the strength to withstand significant pressure and can be made out of molded plastic for disposable use as well as stainless steel and/or machined plastic for reusability. The extraction cartridge can be removed and opened for sample access without the use of tools. The outside of the cartridge can be purged after it is installed in a heated high pressure vessel to remove contamination from its exterior. In one embodiment, the extractor includes a fraction collector fr extractants, an automatic sample changer and an automatic cartridge transfer mechanism which provide completely automated extractions.

摘要: To provide smooth constant flow from a pump, a chromatographic system comprises: a chromatographic column having an inlet; a pump for supplying fluid to the inlet of the chromatographic column; a power means for the pump motor; positive and negative feedback loop means for controlling said power means; means for energizing and de-energizing said positive and negative feedback control means; said negative feedback control means receiving a signal from said means for measuring flow rate and including means for comparing said signal with said corrected flow rate reference signal while said second feedback loop is energized to generate an error signal controlling said power means; and said positive feedback control means applying an acceleration voltage to said motor from a time a preset period after the initiation of a return stroke of said piston until after a timed duration.

摘要: To supply a programmed gradient to a high pressure pump for introduction into a column, a mixer, degasser and accumulator communicates with the high pressure pump to supply fluid thereto, having a volume no more than 150 times the chamber volume of the high pressure pump. The mixer, degasser and accumulator includes temperature sensors which sense when the mixer, degasser and accumulator has been emptied to a level where it cannot respond to a demand for fluid from said high pressure pump and provides a signal to a low pressure pump which responds by cycling to again fill the mixer, degasser and accumulator. Upon receiving a demand signal, the low pressure pump fills by drawing fluid from a plurality of fluid sources to compose the gradient being used at that time, with the pump slowing during valve opening and closing so as to avoid cavitation.

摘要: To reduce tailing, the frit at the outlet to the liquid chromatographic column has a diameter substantially equal to that of the inside diameter of the outlet end of the column and less than two millimeters. It is held directly against the packing of the column within a cylindrical member having a shoulder with a central aperture at least the size of the diameter of the frit and cylindrical walls which are fastened to resist force in the direction of the axis of the column from its inlet to its outlet. A gasket seals the cylindrical holder against the column wall. The frit is formed of gold-plated nickel spheres sintered together at a temperature lower than the gold-nickel eutectic temperature of 950 degrees Celsius but above the phase-immiscibility temperature of 720 degrees Celsius. The size and shape of the frit is selected to reduce tailing by causing the flow path from the column to be uniform.

摘要: To stabilize flow rate in a single-stroke, syringe-type high pressure pump to a constant value after turn-on, a control system increases the pump speed until equilibrium conditions are reached and then pumps at the preset rate. In one embodiment, the control system senses the maximum rate of change of pressure, detects a pressure when the rate of change is two-thirds of the maximum, increases the pump rate until the pressure is three times that at the value of the detected pressure and begins pumping at the preset constant flow rate.In other embodiments: (1) a pumping system is operated at a flow rate ten times the preset flow rate and, when the time derivative of the pressure has dropped to nine tenths of its maximum, the pump motor speed is reduced to the preset flow rate; (2) the outflow of the column is measured under no load conditions at the desired flow rate and when a column is connected, the pumping speed is increased until the same outflow results before reducing the pumping rate, and (3) the system is operated in a constant pressure mode to a point at which the pressure is estimated to provide the desired flow rate, the flow rate measured, and if the flow rate is not the preset one, a new pressure measurement is set and the procedure repeated.
