摘要: An improved analytical method for analysis of dianhydrogalactitol preparations provides a method for determining the purity of dianhydrogalactitol and detecting impurities in preparations of dianhydrogalactitol, as well as identifying any such impurities. The method employs high performance liquid chromatography (HPLC), in particular, HPLC employing a reverse phase amide column with ELSD detection; the HPLC can be followed by tandem mass spectroscopy. The method can further comprise the step of performing preparative HPLC collection of at least one specific substance peak present in a preparation of dianhydrogalactitol.

摘要: Described is a method of generating a flow having a composition gradient such as a mobile phase gradient for liquid chromatography. A pair of pumps is operated such that the initiations of pump strokes for one pump are controlled to occur between the initiations of pump strokes for the other pump so that the sequences of pump strokes fort the two pumps are interspersed in time. Initiations of the pump strokes of the second pump are offset in time relative to initiations of the pump strokes of the first pump such that variations in the flow rates of the first and second pumps due to initiation do not overlap in time. The volume of liquid contributed by a pump stroke is controlled according to the relative contribution of the respective pump to the composition gradient.

摘要: A method of performing a chromatographic separation includes modulating a portion of a flowstream to a trap column to retain at least one analyte from the flowstream on the trap column. A flow of a modifier is provided through the trap column to generate an elution comprising the at least one analyte. A flow of a compressible fluid-based chromatography (CFC) mobile phase or CFC solvent is merged with the elution from the trap column to generate a diluted elution. Carbon dioxide may be used as the CFC solvent or as a component of the CFC mobile phase. The diluted elution is provided to a CFC column where at least one analyte is focused at a head of the CFC column. Examples of a flowstream that may be used include an eluent from a chromatography column or a fluid flow from an extraction system.

摘要: This invention provides methods and compositions, e.g., to reduce interference from non-specific binding sample constituents in a migration shift assay. Interference due to non-specific binding of sample constituents to an affinity substance (e.g., an affinity molecule or a conjugate of an affinity molecule and a charged carrier molecule) is prevented by, e.g., binding the constituents to charged polymers such as heparin sulfate.The present invention also provides methods to concentrate an analyte of interest with high concentration and to detect the analyte with high sensitivity, and further to optimize the reaction conditions for easily concentrating the analyte. Such objects of the present invention are attained, for example, by concentrating a complex of the analyte and a conjugate which is formed by contacting the analyte in a sample with an affinity molecule bound to a charged carrier molecule such as DNA.

摘要: A separation system comprises (i) at least two separation units, each separation unit comprising a fluid inlet and a fluid outlet, wherein the separation units are connected in series outlet to inlet to form a line of separation units, and (ii) sensing and adjustment means, provided in-line between each separation unit, for continuously monitoring and adjusting at least one environmental property parameter of fluid flowing from one separation unit to a subsequent separation unit in the line of separation units. Uses of the separation system and a method for purification of a liquid containing a desired species, using separation units and in-line adjustment of a fluid flow or flows, are also disclosed.
