A procaryotic microorganism and a method for its production is provided wherein the microorganism contains at least one stable foreign DNA portion in the chromosome. The disclosed microorganisms and their progeny are substantially free of genetic rearrangement involving the foreign DNA. In a preferred embodiment, cyanobacteria are employed. The microorganisms are produced by introducing into the cell an insertion vehicle that contains foreign DNA ligated between two portions of DNA homologous to adjacent portions of the recipient's chromosome.

The nucleic acid molecules can be used for directed or localized delivery to tumors or other cells associated with disease resulting in production of a chromophore product that can be used for diagnosis and therapy, including of tumors and metastases.

Diagnostic methods for in vivo and ex vivo detection of oncolytic virus infection of tumors in a subject are provided. The diagnostic methods employ imaging agents that detect macrophage or inflammatory cells, such perfluorocarbon (PFC) imaging agents, to detection of inflammation associated with oncolytic virus administration, and, thereby detect tumors and cancers. Combinations and kits for use in the practicing the methods also are provided.

Provided are methods for using cellular compositions in combination with oncolytic viruses. The methods include administering oncolytic viruses for the inhibition and treatment of tumors caused by administration of cellular therapies, such as stem cell therapies. The methods also include contacting cellular compositions with oncolytic viruses for the removal of neoplastic cells prior to administration of the cellular composition for therapy. Diagnostic methods for monitoring treatment also are provided.

A fusion gene is provided comprising the cDNA of Renilla luciferase and the cDNA of the "humanized" Aequorea green fluorescent protein. The fusion gene was used to produce a novel protein, the "Renilla-GFP fusion protein," which displayed both the luciferase activity of Renilla luciferase, and the green fluorescence of GFP. The Renilla-GFP fusion gene is useful as a double marker for monitoring gene expression quantitatively in UV light and by enzyme activity.
