摘要:
A method for treatment of HIV infection includes administering at least one anti-HIV drug, such as a reverse transcriptase inhibitor, to a patient in need of such treatment and administering an extract from inflammatory tissue inoculated with vaccinia virus to the patient following the administration of the at least one anti-HIV drug. The extract maintains suppressive action on HIV replication, even if the administration of the anti-HIV drug is terminated.

The present invention provides recombinant poxviruses, such as vaccinia virus, that contain an integrated exogenous sequence, such as a foreign gene, encoding a prodrug converting polypeptide that can convert a prodrug to a drug that prevents virus replication or is otherwise toxic to the virus. The recombinant poxviruses can be suitable for use as vaccines. The invention also provides, among other things, methods of inhibiting virus replication, methods of vaccination and methods of treating vaccinated subjects showing signs or otherwise at risk for of vaccination-induced disease.

摘要:
Recombinant poxviruses, such as vaccinia, are provided that comprises a segment comprised of (A) a first DNA sequence encoding a polypeptide that is foreign to poxvirus and (B) a poxvirus transcriptional regulatory sequence, wherein (i) said transcriptional regulatory sequence is adjacent to and exerts transcriptional control over said first DNA sequence and (ii) said segment is positioned within a nonessential genomic region of said recombinant poxvirus. Vaccines, carriers, cells, and media comprising recombinant poxviruses, and methods of immunization with recombinant poxviruses, also are provided.

The Parapox B2L virus envelope protein is used as an adjuvant to enhance a subject's response to an administered antigen. Both antibody and cellular immune responses can be modified. B2L protein is particularly useful as an adjuvant for poorly immunogenic tumor vaccines and subunit vaccines, such as those useful for preventing and/or treating flu, tuberculosis, respiratory syncytial virus, anthrax and HIV.

In this application is described a poxvirus naked DNA vaccine which protects animals against poxvirus challenge comprising IMV and EEV nucleic acids from poxvirus. Methods of use of the vaccine and its advantages are described.

Attenuated recombinant viruses containing DNA coding for a cytokine and/or a tumor associated antigen, as well as methods and compositions employing the viruses, are disclosed and claimed. The recombinant viruses can be NYVAC or ALVAC recombinant viruses. The DNA can code for at least on of: human tumor necrosis factor; nuclear phosphoprotein p53, wildtype or mutant; human melanoma-associated antigen; IL-2; IFN&ggr;; IL-4; GNCSF; IL-12; B7; erb-B-2 and carcinoembryonic antigen. The recombinant viruses and gene products therefrom are useful for cancer therapy.

The present invention involves a recombinant virus which comprises at least one foreign nucleic acid inserted within a non-essential region of the viral genome of a virus, wherein each such foreign nucleic acid encodes a protein. The protein which is encoded is selected from the groups consisting of a feline CD28 protein or an immunogenic portion thereof, a feline cD80 protein or an immunogenic portion thereof, a feline CD86 protein or an immunogenic portion thereof, or a feline CTLA-4 protein or an immunogenic portion thereof. The protein is capable of being expressed when the recombinant virus is introduced into an appropiate host. The present invention also involves a recombinant virus further comprising a foreign nucleic acid encoding an immunogen derived from a pathogen. The present invention also comprises recombinant viruses which are capable of enhancing an immune response in a feline. The present invention also comprises recombinant viruses which are capable of suppressing an immune respons in a feline.

Antigenic virus materials, especially aqueous vaccines of all types in liquid or in freeze-dried form, are stabilized by incorporating therein at least 0.1% by weight of calcium lactobionate. Desirably 2-10% of lactose may also be incorporated and the vaccines of the invention are stated to retain their antigenicity at higher temperatures (e.g. at -20 DEG C. instead of -60 DEG C. for a measles vaccine) and to be able to withstand repeated freeze/thaw cycles. Other vaccines specifically mentioned are poliomyelitis, infectious hepatitis, yellow fever, mumps, rabies, smallpox and vaccinia vaccines.
