Disclosed herein are methods for single-cell sequencing. In some examples, the methods include enriching a sample comprising a plurality of cells for cells of interest to produce an enriched cell sample; isolating one or more cells of interest in the enriched cell sample; and obtaining sequence information of one or more polynucleotides from each of the one or more isolated cells. Obtaining sequence information may include generating a molecularly indexed polynucleotide library from the one or more isolated cells. Enriching the sample may include focusing cells of interest in the sample using acoustic focusing.

Devices for separating magnetically labeled moieties in a sample are provided. Aspects of the devices include a magnetic field source, a first magnetic field guide having a wedge-shaped portion with an apex edge, and a second magnetic field guide having a wedge-shaped portion with an apex edge. The apex edge of the first magnetic field guide is aligned substantially across from and parallel to the apex edge of the second magnetic field guide, and the device is configured to separate magnetically labeled moieties from non-magnetically labeled moieties in the sample. Also provided are methods of using the devices, as well as systems and kits configured for use with the devices and methods.

Abstract:
Aspects of the present disclosure include an acoustic separator for acoustically separating components of a sample, such as a biological sample. Acoustic separators according to certain embodiments include an acoustic field generator soldered to a flow channel having a fluid flow path where the acoustic field generator produces an acoustic field in the fluid flow path. Methods of soldering an acoustic field generator to a flow channel having a fluid flow path are also described. Methods for acoustically separating components of a sample as well as systems and kits, including one or more acoustic separators, suitable for practicing the subject methods are also provided.
