A method for obtaining a virus vector free of a serum, and a serum-free medium for cultivation of a virus producer cell which can be used for the method are provided. By cultivating a virus producer cell using a serum-free medium containing serum albumin, it is possible to cultivate the cell in a state equivalent to that in a serum-containing medium to produce a virus vector at a sufficient titer. The virus vector prepared from the virus producer cell cultivated in the medium exhibits gene transfer efficiency comparable to that of a conventional vector. The medium is useful for gene therapy and studies thereof.

The present invention provides a retroviral vector containing a transcription unit comprising a transcription regulatory sequence and a gene encoding a polypeptide having single-stranded RNA-specific endoribonuclease activity which is placed so that its expression can be controlled by the regulatory sequence, wherein the unit is placed so that the direction of the transcription of mRNA from the unit is opposite to the direction of transcription of the RNA genome of the retroviral vector. By using the vector constructed as described above, viral supernatant showing high gene transfer efficiency can be prepared. The retroviral vector of the present invention is useful for the treatment and/or prevention of cancers and virus infections.

Disclosed is a method for producing a cell population into which a desired gene is introduced, which is characterized by comprising the following steps (1) and (2): (1) culturing a cell population containing a T-cell and/or a progenitor cell of a T-cell in a vessel containing fibronectin or a fragment thereof and a CD3 ligand; and (2) adding a vector carrying the desired gene to the vessel of the step (1). The N-acetylglucosaminyltransferase III activity is enhanced in a cell carrying retrovirus-origin gag-pol gene and env gene. By constructing a retrovirus vector with the use of the above cell, a retrovirus vector having a modified sugar chain structure can be obtained. The retrovirus vector constructed by this method shows a high infection efficiency particularly in the presence of a functional substance.
