    摘要: The present invention provides an automated system and method to isolate nucleated blood cells from whole blood or bone marrow. A disc mounted to a centrifuge system with spinning rotor is used to manipulate cells by channeling fluids while subjected to high gravitational field. The disc embodies at least two axisymmetric processing stations connected by a circular channel. Each station contains multiple chambers connected by fluidic channels to controllably transfer fluids. First stage separation allows for the isolation of the buffy coat layer while the second stage separation utilizes gradient density fluids to isolate the targeted nucleated cells from the buffy coat layer in the spinning disc.

    摘要: A whole blood collection system includes an automated pump/control unit and an accompanying disposable blood set. When combined and connected to a source of anticoagulant, these elements allow automatic priming of the blood set with anticoagulant and automatic collection of anticoagulated blood product according to three different collection modes. The unit's pump and the blood set are specially designed to cooperate during the collection process to assure that the collected product has a precise blood to anticoagulant ratio. During the collection procedure, the pump/control unit automatically collects data relating to the procedure. Additional data specifically identifying components of the blood set, such as the blood collection bag, along with identification data on the donor's registration form may be scanned into the pump/controller unit by a scanner associated with the unit; this facilitates positive sample identification and tracking. At the end of the procedure, a printer in the pump/controller unit automatically prints out this information so that a detailed record is immediately available about the collected blood product, the procedure for collecting it and the source of the product, i.e., the donor.

    摘要: A rotor having variable volumes adapted for collecting and centrifuging biological fluids. The rotor includes an impermeable flexible body having a cylindrical shape with stretchable vertical walls and semi pliant base. The rotor includes a rigid circular member that is seamlessly joined to the top of the flexible body. The rigid cover defines an opening having a rotary seal that maintains a closed system inside a spinning rotor. The rotary seal permits a plurality of non-rotating conduits to pass through for controlling the flow in and out of the rotor while it is spinning. In a preferred embodiment, the rotor includes a Core to stabilize the rotating fluids inside the separation chamber, and/or includes a diverter to divert the fluid entering the rotor to the periphery of the separation chamber for better processing. When the rotor is inserted in the centrifuge, the rigid cover is fixed at the top of the centrifuge bucket. The base of the flexible body is firmly secured to the chuck by vacuum or mechanical interlock means. The chuck moves vertically down and up by pneumatics or electrical motor means embedded in the rotating centrifuge, while it is spinning. The base of the rotor moves vertically in conjunction with the chuck increasing or decreasing the volume of the processing chamber as the sidewall of the flexible body stretches or contracts. In another preferred embodiment the base of the flexible body is secured to the chuck by centrifugal means.
