摘要: Aspects of the invention provide methods for producing one or more secondary metabolites from microbial culture. In various embodiments, the method comprises culturing a microbial cell producing a secondary metabolite for recovery from a bioreactor medium, the medium comprising an aqueous phase and an extraction phase. The composition of the extraction phase, and the relevant amount with respect to the aqueous phase, enhances production of the secondary metabolite from microbial cells and/or enhances extracellular transfer of the metabolite.

摘要: In various aspects, the present invention provides uridine diphosphate-dependent glycosyltransferase (UGT) enzymes capable of catalyzing the transfer of a monosaccharide moiety from a NDP-sugar to the 3′ carbon of a sugar moiety of a substrate, such as a terpenoid glycan, thereby functioning as a “1-3 UGT.” In other aspects, the invention provides polynucleotides encoding the 1-3 UGT, and host cells comprising the same. In still other aspects, the invention provides methods for preparing glycosylated substrates, including steviol glycosides, using the enzyme and host cells of this disclosure.

摘要: The present disclosure in various aspects provides engineered enzymes and encoding polynucleotides, as well as host cells and methods for making rotundone and other terpenoids. For example, in various aspects, the invention provides engineered α-Guaiene Synthase (αGS) and Guaiene Oxidase (GO) enzymes that increase biosynthesis of rotundone from farnesyl diphosphate, and in certain embodiments substantially reduce biosynthesis of side products such as α-Bulnesene or oxygenated side products. In still other aspects, the invention provides engineered terpene synthase enzymes for directing biosynthesis toward a desired product, to thereby improve product profiles and/or product titers from terpene synthase reactions.

When incubating the microbial strain with a plant extract or fraction thereof comprising the intermediates, these glycosylated intermediates are available for further glycosylation by the cell, and the advanced glycosylation products can be recovered from the media and/or microbial cells.
