摘要: Exemplary embodiments eliminate the need for column ovens in serial column chromatography arrangements and systems by using insulated sleeves. The insulated sleeves may encase individual chromatography columns or clusters of chromatography columns. The use of the insulated sleeves allows the chromatography columns to be positioned in close proximity to each other. This may decrease the overall size of a serial column chromatography arrangement and may reduce costs by not requiring the column ovens.

摘要: A system that incorporates teachings of the subject disclosure may include, for example, a process that includes obtaining a porous medium comprising a porous material having a first shape and an initial porosity profile. The porous medium is engaged with a cavity in a fluidic device, wherein the cavity is in fluid communication with a channel of the fluidic device. The first shape of the porous material can be adjusted to a second shape resulting in the initial porosity profile being adjusted to a target porosity profile. Such adjustment can be accomplished by the engaging of the porous medium with the cavity, by pre-adjusting a shape of the porous media before insertion into the cavity, or by some combination thereof. Other embodiments are disclosed.

摘要: The present disclosure discusses a method of separating a sample including oligonucleotides including coating a flow path of a chromatographic system; injecting the sample comprising oligonucleotides into the chromatographic system; flowing the sample through the chromatographic system; and separating the oligonucleotides. In some examples, the coating of the flow path is non-binding with respect to the analyte, such as oligonucleotides. Consequently, the analyte does not bind to the coating of the flow path. The non-binding coating eliminates the need for passivation, which can eliminate the formerly needed time to passivate as well. In addition, analyte can be recovered with a first injection in a system, such as chromatographic system.

摘要: The present disclosure describes methods for analyzing mRNA poly(A) tail sequences with a high resolution to determine poly(A) tail length and heterogeneity. The method involves digesting an mRNA molecule to liberate a poly(A) tail, preparing a chromatographic sample comprising the mRNA poly(A) tails, preparing a second chromatographic sample comprising a reference sequence comprising a mRNA poly(A) tails having a predetermined length, separating the first and second samples by a chromatography method, which result in one or more chromatograms, and determining a sequence length of the mRNA poly(A) tails by comparing the chromatograms of the first and second samples. Chromatography methods for analyzing the poly(A) tail may include ultraviolet size-exclusion chromatography (SEC UV), ultraviolet ion-pair reversed-phase liquid chromatography (IP RP LC UV), ultra high performance liquid chromatography (UPHLC), and combinations thereof.

The cartridge includes a barrel (204) with a first and second end, a column segment (209) connected to the second end of the barrel, and a column (205) containing a sorbent material. The sorbent material includes particles that have antibodies attached to them to selectively retain analytes, proteins attached to them to retain certain classes of antibodies, or enzymes attached to them to perform specific modifications to certain classes of molecules. The column segment can be in thermal communication with a temperature control device in order to control the temperature of the column.

摘要: Described is a method for determining a dwell volume of a liquid chromatography system and a liquid chromatography system that can determined the system dwell volume. The method includes mixing a flow of a first solvent with a flow of a second solvent to form a solvent mixture. The flows of the first and second solvents are decreased and increased, respectively, to generate a gradient composition. A system pressure of the liquid chromatography system is measured to determine a pressure trace defined as the measured system pressure as a function of time. The dwell volume of the system is determined from a time delay determined between the gradient composition at the mixing location and the pressure trace. The method can be performed with a liquid chromatography system having a chromatographic column or a flow restrictor used in place of the chromatographic column.
