摘要:
A nanostructure comprises a MOX NP and a bidentate ligand on a surface of the MOX NP. A cancer recognition molecule is covalent coupled to the surface of the MOX NP via the bidentate ligand. A biocatalyst is also coupled to the surface of the MOX nanoparticle via the bidentate ligand. The cancer recognition molecule includes a structure configured to selectively recognize a corresponding antigen on a surface of a cancer cell and bind to the antigen. The biocatalyst is structured to selectively catalyze the oxidation of a light emitting compound to produce photons. The photons transform the MOX NPs into an excited state such that the MOX NPs generate reactive oxygen species (ROS) in the vicinity of the cancer cells in the excited state. The reactive oxygen species lyse or cause apoptosis in the cancer cells in situ. The biocatalyst includes luciferase and the light emitting compound includes luciferin.

Described are drug carriers useful in magnetic resonance imaging (MRI)-guided drug release comprising a shell capable of releasing an enclosed biologically active agent as a result of a local stimulus, e.g. energy input, such as heat, wherein the shell encloses a 19F MR contrast agent. Preferably, the carrier also acts as a contrast enhancement agent for MRI based on the principle of Chemical Exchange-dependent Saturation Transfer (CEST). To this end the shell encloses a cavity that comprises a paramagnetic chemical shift reagent, a pool of proton analytes, and the 19F contrast agent, and wherein the shell allows diffusion of the proton analytes.

Compositions and methods for assessing inflammation in a subject. The preset disclosure provides compositions for labeling leukocytes with a 19F-containing perfluoropolyether molecule ex vivo. In some examples, the leukocytes are obtained from the patient, enriched in a whole blood fraction, and then labeled. The labeled cells may be re-introduced into the patient. The leukocytes may accumulate at a site of inflammation, thus permitting non-invasive evaluation of inflammation in patients. The present methods provide a tool for assessing inflammation in a wide variety of autoimmune diseases and may have particular utility in intestinal diseases such as Crohn's disease, ulcerative colitis, inflammatory bowel disease, and cardio myositis.

The disclosure generally relates to formation of polymers grafted to or polymerized from the surface of gold nanoparticles. The polymers are functionalized to include therapeutic agents and/or targeting agents at their surface, thereby allowing both therapeutic and targeting compounds to be directed to specific cells in a patient.

The present disclosure is directed generally to lanthanide nanoparticle conjugates, such as gadolinium nanoparticle conjugates, nanoparticle conjugates including polymers, conjugation to targeting agents and therapeutic agents, and their use in targeting, treating, and/or imaging disease states in a patient.

Disclosed herein are nanostructures comprising a polymeric framework comprising at least five geminal bisphosphonate groups, wherein the geminal bisphosphonate groups independently of each other are incorporated as —R3R4C(P═O(OR1)(OR2))2, wherein R1 and R2 are independently selected from the group consisting of a negative charge, H, alkyl and aryl, and wherein at least one of R3 and R4 is a group connected to the polymeric framework with the proviso that when only one of R3 and R4 is such a connected group, the other of R3 and R4 is either a group being able to connect to the polymeric framework, or the residue of such a group, or selected from the group consisting of H, OH, OR5 and R5, wherein R5 is a lower alkyl. The polymeric framework may comprise manganese ions. Disclosed are also methods for producing such manganese containing nanostructures, compositions comprising such manganese containing nanostructures and use of such manganese containing nanostructures, i.a. as MRI contrasting agents.

Disclosed are novel, targeting, paramagnetic nanoparticles as contrast agent for magnetic resonance imaging. The compositions of the nanoparticles are composed of self-assembled polyelectrolyte biopolymers having targeting moieties, which can suitable for targeted delivery of paramagnetic ions complexed to the nanoparticles. The nanoparticulate contrast agent can internalize into the targeted tumor cells to realize the receptor mediated uptake, and therefore afford enhanced relaxivity and improved signal-to-noise effect on the examined tissue areas. Methods for making these targeting MRI contrast agents are also provided.

A marker delivery device includes an elongated delivery cannula which has a distal end section, an inner lumen and a discharge opening in the distal end section in communication with the inner lumen. At least one elongated fibrous marker body is slidably disposed within the inner lumen of the elongated delivery cannula. The at least one elongated fibrous marker body includes a plurality of bioabsorbable polymeric strands. The strands are compressed to a compressed configuration and bound together in the compressed configuration with a polymer binding agent prior to insertion into the elongated delivery cannula. A releasable plug is disposed within a distal portion of the inner lumen and distal to the at least one elongated fibrous marker body so as to at least partially occlude the discharge opening in the distal end section.
