摘要: The present invention relates to systems and methods for sequential operation of staining, imaging and sectioning of tissue samples by a processing system. After each layer of the sample is removed by the sectioning system, the system automatically stains the exposed surface of a sample to a depth to enable imaging of the remaining tissue. The system then repeats the sectioning, staining and imaging steps in sequence to image the sample.

摘要: Systems and methods enable fully automated tissue processing using tissue processing components for producing tissue sections from a tissue sample block and to transfer the tissue sections to slides. A control device queries a Laboratory Information Management System (LIMS) with a sample identifier associated with the tissue sample block and receives tissue sample data from the LIMS. The control device generates tissue processing workflow parameters based on the tissue sample data for each tissue processing component to perform an automated tissue processing workflow. The control device automatically controls each tissue processing component according to the tissue processing workflow parameters to process the tissue sample block. The control device generates a block processing update message while the tissue sample block is processed and communicates the block processing update message to the LIMS to enable the LIMS to track the automated tissue processing workflow for the tissue sample block.

摘要: A kit, including: a first solution, a second solution, and a metal mesh. The first solution includes: dulbecco's modified eagle medium (DMEM), 65-95 V/V %; dimethyl sulfoxide (DMSO), 5.5-20 V/V %; ethylene glycol (EG), 3.5-15 V/V %; bovine serum albumin (BSA), 0.5-4 W/V %; sucrose 1-5 W/V %; methylcellulose with a viscosity of 4000 centipoise (cP), 0.05-0.8 W/V %; hetastarch 0.25-0.6 W/V %; and glucose 15-35 W/V %. The second solution includes: DMEM, 65-95 V/V %; DMSO, 5.5-20 V/V %; EG, 8-20 V/V %; BSA, 0.5-4 W/V %; sucrose, 10-20 W/V %; methylcellulose with a viscosity of 4000 centipoise (cP), 0.05-0.8 W/V %; polyvinyl pyrrolidone (PVP), 0.25-0.6 W/V %; and glucose 15-35 W/V %. The metal mesh has a thickness of 0.15-0.2 mm and includes a plurality of square holes. The side length of the square holes is 2.0-3.0 mm, and the spacing between adjacent holes is 0.5-2.0 mm.

摘要: Methods and reagents are provided for the rapid extraction of nucleic acids from a fixed paraffin embedded sample (e.g., a FFPET sample). In some embodiments, the methods comprise incubating one or more sections of said tissue sample in a lysis solution comprising a buffer sufficient to maintain the pH of said solution at a pH ranging from about pH 4 to about pH 9; a chaotropic agent; a chelating agent; and a detergent; where the incubating is at a temperature ranging from about 50° C. to about 100° C.; and recovering the nucleic acid from said lysis solution.

摘要: The following concerns a method for co-localization of microscopy or histology stains by the assembly of a virtual image from one or more imaging operations. In particular, the method decreases the time required to obtain multiple labeled antigen or protein histology images of a biological sample. The method includes imaging the tissue as it is sliced by a microtome with a knife edge scanning microscope and spatially aligning the samples by the generated images. The spatial alignment of samples enabled by the method allows a panel of different antigen or protein secondary or functional stains to be compared across different sample slices, thereby allowing concurrent secondary stains of tissues and cells.
