A method is disclosed for manufacturing fruit liquor in which muscat fragrance is emphasized. The method includes either or both of set of step (a) and step (d) and another set of step (b) and step (c). Step (a) is to obtain fruit juice or fruit juice containing pericarp, by crushing and squeezing the fruit part of grapes. Step (b) is to heat the fruit juice or the fruit juice containing pericarp obtained in step (a) so as to make the article temperature reach 45 to 95 °C. Step (c) is to add enzyme having the glycosidase activity at the article temperature of fruit juice in a range of 10 to 55 °C. Step (d) is to ferment the fruit juice obtained in step (b) or step (c) using a yeast having the β-glucosidase activity. The fruit liquor manufactured by this method has favorable muscat fragrance.

A polypeptide participating in the biosynthesis of pladienolide as a macrolide compound; a DNA coding for such a polypeptide; a variant thereof; a transformant in which part or the whole of such a DNA or variant thereof has been incorporated; and a process for producing pladienolide as a macrolide compound with the use of such a transformant. In particular, there is provided an isolated pure DNA comprising at least one region coding for a polypeptide participating in the biosynthesis of pladienolide. Further, there are provided a polypeptide coded for by this DNA and a recombinant plasmid capable of self-replication or integration replication carrying this DNA. Still further, there are provided a transformant carrying this DNA and a process for producing pladienolide, characterized in that this transformant is cultured in a culture medium and pladienolide is collected from the culture solution.

A vitamin D 3 hydroxylase is purified from Pseudonocardia autotrophica cell, and a primer is designed based on amino acid sequence obtained from hydroxylase. Subsequently, PCR is conducted using genomic DNA of Pseudonocardia autotrophica as a template to clone a gene for the vitamin D 3 hydroxylase. By conducting a conversion reaction using a microorganism in which the vitamin D 3 hydroxylase gene is expressed using a proper expression system, a hydroxide of vitamin D or the like (e.g., hydroxy vitamin D 3 ) can be produced with high efficiency.

An object is to establish a method of preparation of novel polysaccharides from a cassis polysaccharide (CAPS), wherein the novel polysaccharides exhibit a higher immunoregulatory effect per unit amount, have a low viscosity, and can be handled readily during the method of preparation a final product. Another object is to provide health foods and drinks having high safety and an excellent immunoregulatory effect at a low cost by utilizing a juice, processed juice or a purified product. The present invention relates to a composition of which chief ingredient is novel polysaccharides, having an average molecular weight falling within a range of 10,000 to 40,000; which is obtained by partially digestion of CAPS with enzyme, and an immunoregulatory foods and drinks utilizing the composition.
