Provided herein are methods for transducing a plurality of cells in a composition of cells, such as a population of lymphocytes, containing viral particles. In some aspects, provided methods and reagents for the transduction of cell populations involve binding of agents to a molecule on the surface of the cells. In some cases, the reagents are multimerization reagents and the one or more agents are multimerized by reversibly binding to the reagent. In some aspects, the multimerized agent can provide for transduction and/or expansion or proliferation or other stimulation of a population of cells, and then such agents can be removed by disruption of the reversible bond. Also provided are compositions, apparatus and methods of use thereof.

Adjuvant compositions, vaccines, constructs for preparing the adjuvant compositions and vaccines and methods of using the adjuvant compositions and vaccines to enhance immune responses in subjects are provided herein. In particular, a rapid antibody response to the vaccine including both IgG (in the circulation) and sIgA (mucosal secretory IgA) is elicited. The adjuvants and vaccines may be used for sub-cutaneous or mucosal administration enabling low cost, effective vaccination of subjects. A method of epitope mapping to rapidly identify antigenic epitopes is also provided.

The present embodiments provide for an immunogenic multiple antigen presenting system comprising a polymer to which antigens are associated by complementary affinity molecules. For example, the polymer can be a polysaccharide, or antigenic polysaccharide, to which protein or peptide antigens from the same or different pathogens are indirectly linked. The present immunogenic compositions can elicit both humoral and cellular immune responses to one or multiple antigens at the same time.

Compositions and methods to utilize coat proteins of tobacco mosaic virus (TMV) as a scaffold with commonly available linker chemistries and surface antigens to enhance immune responses when used as a vaccination; randomized library to introduce a reactive lysine or cysteine residue externally located at the amino-terminus of the coat protein. TMV coat protein modification allows a controlled stoichiometric biotinylation of the Introduced lysine or cysteine residues further allowing the attachment of antigens using streptavidin as a linker to the biotinylated coat protein. A TMV-protein complex vaccine prepared utilizing this invention results in enhanced immunogenicity and a detectable humoral immune response after only a single immunization in mice.

The reagents comprise conjugates of a first binding member specific for a target in a mammalian host, such as a toxin, drug of abuse, microbe, autoreactive immune cell, infected or tumourous cell, antigen presenting cell, or the like, joined to a second binding member specific for along-lived blood component, including cells, such as an erythrocyte, platelet or endothelial cell, and plasma proteins. These conjugates find use by extending the lifetime and availability of the target binding member for coupling the target and the blood component and thereby reducing the concentration free target, modulating the volume of distribution of the target, targeting the target to sites of enhanced immune response, facilitating target clearance from the bloodstream, or extending the stimulation of an immunogen.
