Also provided are methods of preparing a Fn3 polypeptide monobody, and kits to perform the methods.

摘要: PROBLEM TO BE SOLVED: To provide a method for identifying molecules which mimic one idiotype of an autoimmune disease-associated auto-antibody (autoantibodies).
SOLUTION: The method comprises the following steps: (a) purifying autoantibodies from sera of one or more patients suffering from the autoimmune disease; (b) forming an affinity matrix by binding the autoantibodies to a solid phase; (c) bringing pooled plasma or B cells comprising immunoglobulins into contact with the affinity matrix, subsequently removing unbound plasma components; (d) eluting the bound immunoglobulins which are anti-idiotypic antibodies (anti-Id) to autoantibodies from the matrix; (e) providing a molecular library comprising a plurality of molecule members; and (f) bringing the anti-Id into contact with the molecular library and isolating those bound molecules bound with the anti-Id (the bound molecules are those imitating one idiotype of the autoimmune antibodies).
