摘要: A flow path comprising a reaction cavity in which there is a solid phase to which an affinity reactant I1 has been immobilized by the use of an immobilizing pair of reactive structures that comprises a) a plurality of functional equal structures RSsp on the solid phase, and b) a structure RSari on affinity reactant I1, where RSsp and RSari are mutually reactive with each other to the formation of a link structure that immobilizes affinity reactant I1 to the solid phase. The characteristic feature is that the solid phase comprises a plurality of structures that derive from RSsp but do not immobilize affinity reactant I1 to the solid phase.

摘要: A method for determination of an analyte in an original liquid sample by performing an inhibition affinity assay in a microchannel structure of a microfluidic device. The main characteristic feature is the steps of: (i) providing the microfluidic device in a form where the microchannel structure comprises a reaction microcavity which a) an inlet end, and b) contains a solid phase with binding sites (BS) for an analyte or an analyte-related entity, (ii) providing a liquid sample (1) containing the analyte or the analyte-related entity within the microchannel structure at the inlet end of the reaction microcavity, and flowing sample (1) through the reaction microcavity under conditions such that the analyte or the analyte-related entity is captured by BS leaving a portion of BS unoccupied (free BS), (iii) providing a liquid sample (2) which contains an analytically detectable analogue of the analyte or of the analyte-related entity, and flowing sample (2) through said reaction microcavity to capture the analogue by free BS, and (iv) measuring the amount of analyte analogue captured in step (iii) and relating this amount to the amount of analyte in an original sample.

摘要: The invention is a method for grading a polyclonal antibody response with respect to a desired 5 binding specificity. The response is provoked by an antigenic agent in a host, preferably a vertebrate host. The characteristic feature of the method comprises the steps of: (i) providing a reaction microcavity having an inlet end and an outlet end and containing a solid phase that exposes an immobilized binding structure (BS) that are capable of affinity binding antibodies (Ab) of the response, (ii) flowing a liquid sample containing antibodies of the response in the direction from the inlet end to the outlet end through the solid phase, (iii) determining the distribution of antibodies, which are captured during step (ii), along the flow direction of the solid phase, and (iv) grading the response based on the distribution determined in step (iii).

摘要: A microfluidic device that comprises a microchannel structure in which there are one, two or more flow paths (101;201a,b;301a,a′,b) all of which comprises a porous bed I (104,204,304) that is common for all of the flow paths and exposes an immobilized reactant R that is capable of interacting with a solute S that passes through the bed. The characteristics are that at least one of the flow paths comprises/comprise a second porous bed II (105,205,305) that is placed upstream of porous bed I (104,204,304) and is dummy with respect to interaction with solute S but capable of interacting with a substance DS that is present in a liquid aliquot together with solute S and is capable of disturbing the result of the interaction between solute S and said immobilized reactant R. There is also disclosed a method utilizing the device and variant of the device in which the immobilized R is replaced with a generic affinity ligand LI and/or porous bed II exposes a generic ligand LII that may be different from LI.
