Abstract: The present set of embodiments relate to a system, method, and apparatus for separating cells from a suspension using a gravity driven cell separation device. The apparatus may include a chamber that is separated into a gas compartment and a liquid compartment allowing for transfer of gas through a membrane into the liquid compartment where the cell suspension flows during operation. The embodiments are designed to maximize the gravitational separation effect on the suspension while mitigating the loss of surface area through which gas transfer can occur through a membrane to provide nutrients to cells in the suspension by tilting the cell separation device on two or more axes.

Abstract: Systems and methods of providing run-time quality control and monitoring of a single or multiple sequencing runs are provided herein. In some embodiments, the run-time system includes or is in communication with a processor capable of determining various types of run-time information relating to the quality, progress, etc. of various sequencing runs. In some embodiments, the system can also be in communication with a user interface, for example, a GUI, capable of representing and communicating various types of information to a user regarding the quality of the individual or multiple runs, the functioning of the instrument, an error event, etc. Additionally, the system can capable of receiving actionable information from a user via the GUI thereby allowing the user to terminate or repeat various sequencing steps in a particular run, terminate a entire run, terminate all runs, allow a run to proceed, etc.

Abstract: Various embodiments of a fluidic system of the present teachings are configured to execute a sequence of fluidic operations over the course of a next generation sequencing analysis for the sequential delivery of various solutions used over the course of analysis to a multilane sensor device. Exemplary fluidic operations include washing, priming and nucleotide reagent delivery through a fluidic multiplexer block that is configured to provide independent fluid distribution to each lane of a multilane sensor device used for detection during an analysis. Accordingly, any number or combination of lanes can be used during an analysis, so that during an analysis one lane in any position can be used singly during a run, all four lanes can be used simultaneously during a run, or any combination of lanes can be used simultaneously during a run.

Abstract: A method of aligning a plurality of targets is provided. The method includes generating a plurality of targets. A third phase includes the plurality of targets. The method further includes combining a first phase, a second phase, and the third phase in a volume. The first phase, the second phase, and the third phase are substantially immiscible, and the third phase is in fluid communication with the first phase and the second phase, and the first phase, the second phase, and the third phase are operable to be in a configuration of the third phase between the first phase and the second phase in the volume.

Abstract: A buffered suspension includes a surfactant and a solid buffer particulate having a point of zero charge at least 1.2 pH units different that the pH of the buffered suspension. The buffered suspension can be prepared by mixing a stock solution with the solid buffer particulate and titrating. A method of preforming a pH sensitive process includes drawing the buffered suspension from a reservoir, filtering the solid buffer particulate from the buffered suspension, and applying the filtered solution to a sensor.
