Abstract: A microscope for computational microscopic layer separation may include an imaging device that includes a lens and an image sensor, an illumination system for illuminating a sample, and an actuator to adjust an axial position of a focal plane with respect to the sample. The microscope may also include a processor operatively coupled to the imaging device and the illumination system. The processor may be configured to measure, using the image sensor and the illumination system, optical aberrations of the imaging device at the axial position, and determine whether to adjust the focal plane with respect to the sample in response to the one or more optical aberrations. Various other systems and methods are also disclosed.

The processor may be configured to execute instructions which cause the microscope to capture, using the image capture device, an initial image set of the sample, identify, in response to the initial image set, an attribute of the sample, determine, in response to identifying the attribute, a three-dimensional (3D) process for sensing the sample, and generate, using the determined 3D process, an output image set comprising more than one focal plane. Various other methods, systems, and computer-readable media are also disclosed.

Abstract: Systems and methods are disclosed for constructing an image of a sample using a plurality of images acquired under multiple illumination conditions. In one implementation, a microscope may include an image capture device, an illumination assembly, and a controller. The controller may cause the illumination assembly to illuminate the sample at a series of different illumination conditions. The controller may further acquire from image capture device a plurality of images of the sample, wherein the plurality of images includes at least one image for each illumination condition. The controller may also determine, from the at least one image, image data of the sample for each illumination condition. The controller may further generate, from the determined image data for each illumination condition and in a non-iterative process, a reconstructed image of the sample. The reconstructed image may have a second image resolution higher than the first image resolution.

A method of preparing a microscope slide for imaging a sample on the microscope slide may include placing a transparent flowable material on the sample while the slide is supported with one or more linkages coupled to a processor, covering the sample with a coverslip, and placing the microscope slide on a microscope stage with the one or more linkages coupled to the slide and the processor. Abstract: A microscope including an illumination assembly, an image capture device and a processor can be configured to selectively identify regions of a sample including artifacts or empty space. By selectively identifying regions of the sample that have artifacts or empty space, the amount of time to generate an image of the sample and resources used to generate the image can be decreased substantially while providing high resolution for appropriate regions of the computational image. The processor can be configured to change the imaging process in response to regions of the sample that includes artifacts or empty space. The imaging process may include a higher resolution process to output higher resolution portions of the computational image for sample regions including valid sample material, and a lower resolution process to output lower resolution portions of the computational image for sample regions including valid sample material.

Abstract: Microscopes and methods for processing images of a sample are disclosed. In one implementation, a microscope includes an illumination assembly configured to illuminate the sample under two or more different illumination conditions. The microscope further includes at least one image capture device configured to capture image information associated with the sample and at least one controller. The at least one controller is programmed to receive, from the at least one image capture device, a plurality of images associated with the sample. At least a first portion of the plurality of images is associated with a first region of the sample, and a second portion of the plurality of images is associated with a second region of the sample. The at least one controller is further programmed to initiate a first computation process to generate a high resolution image of the first region by combining image information selected from the first portion of the plurality of images; receive, after initiating the first computation process and before completing the first computation process, a request associated with prioritizing a second computation process for generating a high resolution image of the second region; and initiate, after receiving the request, the second computation process to generate the high resolution image of the second region by combining image information selected from the second portion of the plurality of images.
