    摘要: Cells useful in the pharmacological characterization of inhibitors or activators of proteins are provided. These cells a protein, the presence of which affects a particular characteristic of the cell which changes in response to substances which inhibit or activate the protein. Such responsive changes in a phenotypic characteristic may be utilized in many useful ways, including the discovery, development or characterization of substances suitable for the treatment of diseases or other conditions in human beings or animals. Such cells may also be useful for studying diseases or other biological processes, for determining the effects of various drugs alone or in combination, as well as for identifying or characterizing substances which may be useful in reducing or preventing the occurrence of a disease or other condition. Through the use of such cells in a cell based assay, tamoxifen is identified as an inhibitor of Protein Kinase C (PKC) activity in cell culture.

    摘要: A method and apparatus for electrochemical detection of analyte in a sample makes use of a binding interaction and relies on the discovery that asymmetric distribution of a redox enzyme between two electrodes that occurs when a redox enzyme-containing reagent is immobilized at the surface of one electrode can be detected as a chemical potential gradient arising from an asymmetry in the distribution of oxidized or reduced redox substrate. This chemical potential gradient can be detected potentiometrically by observing the potential difference between the electrodes in an open circuit, or amperometrically by observing the current flow between the electrodes when the circuit is closed. In both cases, the observation of asymmetry can be done without the application of an external potential or current to the electrodes.

    摘要: The present invention provides an ex vivo method for aiding the diagnosis of Alzheimer's disease in a patient comprising: (i) determining the number of alleles of ApoE4 in the patient's genome; (ii) determining the combined expression level of at least three platelet proteins in a platelet sample from the patient; and (iii) comparing the resulting value of step (ii) to a control value, wherein the at least three platelet proteins include at least one isoform of alpha-tropomyosin containing exon 1a and at least two platelet proteins selected from monoamine oxidase-B, coagulation factor XIIIa, wild-type GSTO-1 or mutant GSTO-1, wherein a result higher than the control value is indicative of Alzheimer's disease.The invention also provides a solid support comprising one or more ligands of at least one isoform of alpha-tropomyosin containing exon 1a, and one or more ligands of at least two platelet proteins selected from monoamine oxidase-B, coagulation factor XIIIa, wild-type GSTO-1 protein and/or mutant GSTO-1 protein immobilized thereon.
