The invention provides isolated nucleic acid molecules encoding polypeptides having 2,5-DKG permease activity, and oligonucleotides therefrom. The isolated nucleic acid molecules can be expressed in appropriate bacterial cells to enhance the production of 2-KLG, which can subsequently be converted to ascorbic acid. Further provided are isolated polypeptides having 2,5-DKG permease activity, immunogenic peptides therefrom, and antibodies specific therefor. The invention also provides methods of identifying novel 2,5-DKG permeases.
Abstract:
A system and method for managing data resources includes a policy definition layer, a policy deployment layer and a policy execution layer. The policy definition layer provides for creating, editing and visualizing policies through a user interface using intuitive simple language constructs. The policy deployment layer converts the created policies into a relational format that can be directly stored in a policy database. The policy execution layer executes the deployed policies on occurrence of an event. A resource abstraction and notification layer provides the interfacing of the resources with the policy definition layer, policy deployment layer and the policy execution layer.

Disclosed herein are methods for detecting a water leak in a locomotive engine cooling system comprising numerous components, each being a potential source of leakage of water. The method typically comprises performing at least one inspection providing an indication of a water leak from an engine cooling system of the locomotive. The step of performing at least one inspection may comprise at least one of several steps including performing a visual inspection to determine if water is leaking from a portion of the cooling system external to the engine; performing a visual inspection of exhaust of the engine to detect an indication of water vapor in the exhaust; performing a visual inspection to detect an indication of water leaking into an intake air manifold 30 of the engine; performing an engine oil analysis to detect water in the engine lubricant; reviewing a fault log history for the locomotive; and checking a level of a cooling system sight glass. The method allows for the isolation of potential sources of the water leak from one another so as to prevent false positive leak identification caused by transient water flow between the potential sources. If an engine cylinder assembly is a component identified as a potential source of the water leak during the water pressurization test, the method enables further testing to confirm leakage at such cylinder assembly, before undertaking the costly and time-consuming process of removing the cylinder assembly from the engine. Cylinder assemblies can be tested for leaks by performing an air leak test disclosed herein.

A client/server networking topology comprising a trusted server agent (TSA) (e.g., software application) that runs either at a client node or at a node in close proximity to the client and provides various trusted services to the client on behalf of a trusted server. In instances where the node in which the TSA software runs (i.e., the client or a node in close proximity) may be untrusted, and/or unreliable, methods are provided for making the TSA software running on the untrusted and/or unreliable node provide trusted and reliable services. In one aspect of the invention, a method for providing trusted service in a client/server system comprises the steps of: providing at least one client; providing at least one trusted server (TS); providing at least one trusted server agent (TSA) which is executing on or near the at least one client; providing a trusted service by the at least one TSA to the at least one client on behalf of the at least one TS.
