    摘要: It was found that a fragment of native Thermus aquaticus DNA polymerase (TaqWT) lacking 288 N-terminal amino acids (TaqΔ288) possesses an increased thermostability over TaqWT, TaqΔ279, and TaqΔ289. The present invention therefore provides TaqΔ288, recombinant expression vectors encoding the same or derivatives thereof, as well as purification protocols for TaqΔ288. The invention also encompasses kits containing TaqΔ288 as well as the use of TaqΔ288 and kits containing TaqΔ288. In addition, the invention encompasses methods for the sequencing a nucleic acid template and methods for amplifying a target nucleic acid.

    摘要: Described is a method for the detection of a RNA molecule, the method involving the steps of providing a sample containing the RNA molecule; hybridizing to the RNA molecule a first polynucleotide; extending the first polynucleotide to generate a first strand cDNA; hybridizing a second polynucleotide to the first strand cDNA; extending the first strand cDNA to generate an extension reaction product; amplifying the extension reaction product by means of polymerase chain reaction; and detecting the amplification product by means of real-time fluorescence readout. Also described is a kit containing a first and a second polynucleotide as defined in the present invention, a set of dNTPs, a reverse transcriptase enzyme, and a detection moiety.

    摘要: The current invention restates substituted nitroindole nucleosides as both terminal as well as internal building blocks of labeled oligonucleotide probes for the detection, analysis and quantitation of nucleic acids. The substituent comprises a linker and a detectable group or a linker and a reactive group for post synthesis coupling. These modified nucleosides grant access to a wide application area. These new substituted nitroindole nucleosides can be used as labeling reagents for the facile preparation of, e.g., optimized hybridization probes, simple probes, TAQMAN-probes or molecular beacon probes.
