More specifically, the invention describes methods wherein beta-glucan is concentrated from bran, whole grain and endosperm flours through various slurrying steps in a high concentration alcohol media utilizing various combinations of enzyme and alkali treatment steps.

Chitosan is a natural product having wide range of applications in the food and cosmetic industries. Food and commodity grade chitosan are laden with pyrogens, such as endotoxins and proteins which limit its applicability in the biological and medical arenas, as minute amounts of endotoxins may induce adaptive and innate responses when contacted with mammalian tissue, pharmaceuticals and biomedical devices. Due to chitosan's ability to avidly bind endotoxin and other pyrogens, they are difficult to remove. The present invention is directed to methods for purifying chitosan from shells, food and commodity grade chitosan into ultra-pure, low endotoxin chitosan having biological and medical applicability. Additionally, the present invention is also directed to a method of determining the pyrogenicity of the ultra-pure low endotoxin chitosan.

The present invention provides an efficient and improved process for the production and purification of high molecular weight hyaluronic acid (HA). One embodiment relates to providing growth conditions for production of high molecular weight HA in Streptococcus zooepidemicus. For example, growth in medium comprising a lower protein concentration, e.g., 1% casein hydrolyzate, and a higher sugar concentration, e.g., 5% sucrose, gave 5-6 g/L of high molecular weight HA in the range of 3.5-4.0×106 Da. The present invention also provides an efficient process for the purification of HA comprising treatment with silica gel then active carbon, and subsequent diafiltration with less volume of solvent.

The basic boronate resin can bind about 50% of the monosaccharides and about 10% of oligosaccharides in the basic oligosaccharide solution, by weight of total carbohydrates therein.

A process for reducing the protein content and preserving the capsular polysaccharide content in a complex cellular Streptococcus pneumoniae lysate broth prior to purification is described. Utilizing pH reduction after cellular lysis has resulted in a purified polysaccharide that consistently meets the protein specification, and higher recovery yields of polysaccharide during the purification process.

A novel method to extract and purify cereal β-glucan is described. Cereal β glucans are distinctive polymers of glucose differentiated from other polymers not only by their source but also by their physicochemical properties. The high purity of the cereal β-glucan obtained according to the present invention allows for the preparation of clear, colourless viscous liquid preparations. These liquid preparations are stable to gelling effects when kept at ambient temperatures and low ash concentrations. Compositions comprising β (1-3) β (1-4) glucan and a freezing point depressant are also, described.

Processes are disclosed for the purification and recovery of polysaccharide gums from an aqueous solution, particularly xanthan gum from a fermentation broth. An aqueous solution of at least one polysaccharide gum is mixed with a non-solvent stream comprising water and a subprecipitant level of a non-solvent of the polysaccharide gum. The mixture is concentrated to increase the polysaccharide gum concentration, and optionally undergoes a heat treatment. Additional non-solvent is added to the concentrated mixture to precipitate the polysaccharide gum. The precipitated gum is dried after being separated from the liquid component of the mixture. The removed liquid component can be recycled to the earlier step in the process in which the polysaccharide gum solution is mixed with the non-solvent stream.
