摘要: Method for producing YY super-male and XY physiological female common carps, with which YY-chromosome super-male common carp can be cultivated and androgenetic YY super-male common carp is produced, where microsatellite markers are used for paternity testing and test crossing confirmation. The YY common carp is crossed with normal female common carp to produce the progenies of only male common carp, and without sex identification, the juvenile male common carp with known sex are subjected to artificial sex reversal to produce XY physiological female common carp.

摘要: The invention discloses a method for controlling fish reproduction, the method comprises the following steps of: A, constructing a recombinant gene CMV-eGFP-SV40-CMV-Ga14-SV40 and establishing a GAL4 transgenic zebrafish line; B, constructing a recombinant gene CMV-RFP-SV40-UAS-antisense dnd and establishing a uas-antisensednd transgenic zebrafish line; and C, hybridizing by taking homozygous transgenic fish of the two lines as parents, and the hybrid generation fish was sterility. The method of the present invention takes a strategy that two fertilizable lines hybridized but hybrid generation is sterility, establishing a novel method with a universal meaning for controlling fish fertility; The method solves the contradiction of the reproductively sterility of fish and the hereditability of characters in a reproduction operation research and can be effectively applied to cultivation of new fish specie and population control.

摘要: A medium for culturing Balantidium ctenopharyngodoni in vitro, method for preparing the medium and method for culture in vitro are provided, which belongs to technical field of in vitro culture of intestinal protozoa. The formulation of the culture medium includes: 100 ml of Ringer's solution, 0.5 g of yeast extract, 1.0 g of proteose peptone, 3-6 ml of fetal bovine serum, 6-10 ml of horse serum, 300-500 μl of Bacillus licheniformis suspension, 200-300 mg of aseptic starch. Culture steps include: inoculating collected Balantidium ctenopharyngodoni into a prepared medium, filling with nitrogen and sealing, culturing at 15° C. for 48-72 hours, then transferring to another fresh medium, cycling back and forth, proliferating the Balantidium ctenopharyngodoni continuously. Balantidium ctenopharyngodoni are capable of achieving cell division and proliferation in the culture medium, and lays foundation for the physiological and experimental ecology research of the Balantidium ctenopharyngodoni.

摘要: A test device for noninvasive testing of fish habitat selection and application thereof is described herein. The device includes a camera device, a water circulating system, a fish transporting barrel, and a test water tank. The fish transporting barrel is a water barrel with an openable bottom. The central area of the test water tank is arranged as a closed adaptation area, a water outlet is formed in the bottom of the adaptation area. Fish passages are formed in the adaptation area. Test areas with equal sizes are formed around the adaptation area through partition plates. A water inlet is formed in each test area. Water flow buffering plates are arranged corresponding to the water inlets. Fish passages are formed in the bottoms of the partition plates of the test areas. The water inlets of the test areas are connected to the water circulating system.

摘要: The present application disclosed a method for preparing a sterile transgenic fish, comprising constructing antisense RNA expression vector of salmon-type gonadotropin-releasing hormone, introducing the recombinant DNA fragment into carp oosperm by microinjection, and screening the sterile transgenic fish by Polymerase Chain Reaction and radioimmunoassay, wherein the expression vector comprising a promoter of carp beta actin (β-actin) gene, a complementary DNA fragment of antisense salmon-type gonadotropin-releasing hormone (sGnRH) gene from carp with 323 bp as a target gene comprising sGnRH decapeptide, the coding region of gonadotropin-releasing hormone associated peptide and 3′ non-coding sequence, and 3′ flanking sequence of grass carp growth hormone gene as a stop sequence. The method of the present invention is easy and convenient for operation which provides a basis for providing a technical platform of general applicability for solving the hereditary and ecological safety problems of transgenic fishes.
