Abstract: Provided herein are methods of engineering a reproductive barrier in an insect including (a) introducing into a first insect a first nucleic acid sequence, where the first nucleic acid sequence targets a genomic sequence, where the genomic sequence is proximal to a transcription start site of a gene product; and (b) introducing into a second insect a second nucleic acid sequence encoding a nuclease-deficient endonuclease; and (c) genetically crossing the first insect with the second insect, where the second insect includes a second copy of the genomic sequence where the second copy of the genomic sequence is proximal to a transcription start site of a second copy of the gene product.

Abstract: A method of making sterile diploid organisms includes mating a first population and a second population of single knock-in diploid organisms, wherein the first population of single knock-in diploid organisms are heterozygous organisms expressing a first marker inserted into a gene required for fertility, wherein the second population of single knock-in diploid organisms are heterozygous organisms expressing a second marker inserted into the gene required for fertility, wherein introduction of the first/second marker disrupts expression of the required fertility gene creating a first/second mutant allele of the gene required for fertility, and wherein the first and second markers are distinct; sorting offspring produced from the mating based on their expression of the first and/or second markers; and isolating the sterile diploid organisms, wherein the sterile diploid organisms are heteroallelic diploid organisms expressing the first marker in the first mutant allele and the second marker in the mutant second allele.

Abstract: Four zebrafish gene promoters, which are skin specific, muscle specific, skeletal muscle specific and ubiquitously expressed respectively, were isolated and ligated to the 5′ end of the EGFP gene. When the resulting chimeric gene constructs were introduced into zebrafish, the transgenic zebrafish emit green fluorescence under a blue light or ultraviolet light according to the specificity of the promoters used. Thus, new varieties of ornamental fish of different fluorescence patterns, e.g., skin fluorescence, muscle fluorescence, skeletal muscle-specific and/or ubiquitous fluorescence, are developed.

Abstract: This application relates to methods and compositions for inducing immune tolerance. Specifically, methods and uses of regulatory T cells (Treg) and associated compositions for the induction of immune tolerance are described. The methods and uses may be used to prevent transplant rejection, and in the treatment of diseases or conditions such as graft versus host disease, autoimmune disease and allergies.
