The present invention relates to mutants of lactic acid bacteria which are resistant towards the antibiotic D-cycloserine and/or functionally equivalent antibiotics and which were found to give an increased texture when grown in milk while maintaining the other growth properties of the parent strain. The present invention, furthermore, relates to compositions comprising such mutants, and to dairy products fermented with the lactic acid bacteria resistant towards D-cycloserine and/or functionally equivalent antibiotics.

Novel food-grade cloning vectors comprising a nonsense mutation suppressor-encoding gene, which vector, when it is present in a lactic acid bacterial strain, permits such a strain to have an industrially appropriate growth rate and metabolic activity. The cloning vectors are useful when present in lactic acid bacteria used as starter cultures in the preparation of food or feed products, or a dairy flavor.

Methods for the isolation of mutant rhizobial strains with improved competition of nodulation (Comp.sup.+) is presented. Mutants are selected for constitutive expression of inducible nod genes (in the absence of inducer) and screened for hyperinduction of nod genes in the presence of inducer. Mutants which are defective in nodulation or in symbiotic nitrogen fixation are eliminated from further testing. Selections for constitutive nod expression and for hyperinduction of nod genes are facilitated by the use of plasmid carrying a selectable marker fused to an inducible nod gene and a plasmid carrying a reporter gene fused downstream of an inducible nod gene. The methods exemplified are particularly useful for the isolation and identification of Comp.sup.+ mutants of Bradyrhizobium japonicum.
