    摘要: The present invention relates to a high throughput targeted proteomics-based method for identifying and quantifying one or more binding molecules in a biological fluid of an individual. The present invention involves the use of a plurality of bait molecules to capture the binding molecules. The invention uses proteomics techniques, such as mass spectrometry, for the identification and quantification of the binding molecules. The methods of the invention can be used to identify and quantify antibodies produced by an individual, and proteins expressed by an individual, following exposure to antigens, such as viral antigens, particularly coronavirus antigens and more particularly SARS-CoV-2. Exposure may be via vaccination or following natural infection. The methods of the invention can be used to identify and quantify molecules which bind to a range of proteins of interest, such as autoantigens and neoantigens, as well as to viral vectors.

    摘要: A membrane-spanning actuatable nucleic acid nanopore is provided. The nanopore comprises: one or more polynucleotide strands that provide a scaffold component and a plurality of polynucleotide strands that provide a plurality of staple components wherein each of the plurality of staple components hybridise to the scaffold component; and a trigger that is actuatable in response to a stimulus. Actuation of the trigger results in a detectable conformational change of the nanopore from a first conformation to at least a second conformation. Detection may be via a signal change, such as via fluorescence (e.g. FRET) or a change in an electrical signal readout. Semi-fluid membranes, and sensor devices comprising the actuatable nucleic acid nanopores are also provided.

    摘要: Examples of the present disclosure relate to a particle beam dose profile measurement apparatus comprising a particle detector stack comprising a plurality of scintillator layers. Each scintillator layer of the detector stack is disposed along an axis of the apparatus such that the axis projects through each layer. Each scintillator layer is configured to produce scintillation light indicative of an energy deposition, in that scintillator, of a particle beam incident upon the detector stack along said axis. The apparatus comprises readout circuitry configured to measure the scintillation light of each scintillator layer; and dose profile determination circuitry configured to determine a dose profile of said particle beam within the detector stack. Said determining is based on the measured scintillation light of each scintillator layer, and a quenching correction.

    摘要: The present invention relates to assays for predicting the presence, absence or development of CIN3 and/or cancer, particularly cervical or endometrial cancer, most preferably cervical cancer, in an individual, particularly cervical and endometrial cancer, by determining the methylation status of certain CpGs in a population of DNA molecules in a sample which has been taken from the individual, deriving an index value based on the methylation status of the certain CpGs, and predicting the presence, absence or development of CIN3 and/or cancer, particularly cervical or endometrial cancer, most preferably cervical cancer, in the individual based on the cancer index value. The invention further relates to a method of treating and/or prevention of cancer in an individual, particularly cervical and endometrial cancer, the method comprising assessing the presence, absence or development of CIN3 and/In or cancer, particularly cervical or endometrial cancer, most preferably cervical cancer, in an individual by performing the assays of the invention, followed by administering one or more therapeutic treatments or measures to the individual based on the assessment. The invention further provides a method of monitoring the CIN3 and/or cancer status of an individual according to changes in the individual's cancer index value over the course of time. The invention further relates to arrays which are suitable for performing the assays of the invention.
