It is an object of the present invention to provide an expression vector, a host, a fused protein, a protein, a process for producing a fused protein, and a process for producing a protein, which can prevent formation of an unactive abnormal protein at production of a recombinant protein, and can produce a desired protein as a natural type, that is, a soluble type at a large amount and effectively. That is, the present invention is an expression vector, which comprises: (a) a first coding region encoding a polypeptide having molecular chaperone activity, and (b) a region having at least one restriction enzyme site in which a second coding region encoding a protein can be inserted. In the expression vector of the present invention, the first coding region is operatively linked to a promoter, and the restriction enzyme sites are in the same reading frame as the first coding region, and are downstream of the first coding region, or the restriction enzyme sites are disposed so that the inserted second coding region is operatively linked to a promoter, and the first coding region is in the same reading frame as the second coding region, and is downstream of the second coding region.

The chaperonin-target protein complex in the present invention includes a fusion protein having a chaperonin subunit and an affinity tag linked to the chaperonin subunit via a peptide bond and a target protein for which the affinity tag shows a specific affinity, wherein the target protein is bound to the affinity tag by means of the specific affinity, thereby forming a chaperonin ring structure consisting of a plurality of chaperonin subunits. The chaperonin-target protein in the present invention stabilizes the target protein and surely immobilize on a carrier without causing any change in its stereostructure.

It is intended to provide an immunogen and a composition for immunological use by which a sufficient immune response is induced even by an antigen protein which has been considered unavailable to induce any effective immune responses for various reasons, and a method of producing an antibody using the same. The immunogen containing a fusion protein including the full-length or a part of a desired antigen protein and a folding factor or its subunit linked thereto via at least one peptide bond. Owing to the function of the folding factor, the antigen protein contained in the fusion protein is obtained as a soluble protein having a correct stereostructure. It is also possible to protect the antigen protein and from rapid decomposition in the blood of an animal having been inoculated therewith. The immunogen induces a higher immune response for an animal than an antigen protein alone. Examples of the folding factor include chaperonins and PPIases.

This invention provides a recombinant protein expression system using a host and cell-free translation system, and is capable of universally expressing a large amount of any protein as soluble protein, while preventing toxicity in hosts, formation of inclusion bodies, and decompositions with proteases. Such may be achieved by expressing the desired protein as a fusion protein with chaperoning, such as about 60 kDa molecular chaperons, 60 kDa heat shock proteins, or thermosomes, and accommodating the desired protein inside of a stereostructure of a chaperonin. The present invention provides a process for producing a protein, which comprises transcribing and translating a gene containing a gene encoding the linked chaperonin subunits and a gene encoding a desired protein thereby synthesizing a fusion protein having the desired protein linked via a peptide linkage to the linked chaperonin subunits.

The purpose of the present invention is to provide methods for producing recombinant polyclonal immunoglobulins using recombinant DNA techniques, which provides constant supply of immunoglobulin preparations with minimum risk of infection. It is an object of the present invention to a method whereby a humoral immune response is induced more efficiently in producing an antibody against an antigen protein by gene immunization. A fusion gene composed of a gene encoding the full-length of a part of the antigen protein or a gene encoding a chaperonin subunit or a chaperonin subunit linkage linked thereto is administered to express the fusion gene in the animal, thereby inducing a humoral immune response to an antigen protein by administering. An example of the chaperonin includes Escherichia coli GroEL. There is also provided with a composition for immunization, a method for producing an antibody, a method for producing a hybridoma, and a method for producing a monoclonal antibody.
