摘要: Compositions and methods are provided that are useful for predicting and controlling the stability of expressed polypeptides. The compositions and methods may be used to predict and as desired, increase or decrease the stability of proteins recombinantly expressed in mycobacteria, for example DesA3 expressed in Mycobacterium smegmatis. At the C terminus and the penultimate position, substitution to residues with charged side chains, large non-polar side chains, or no side chains can be used to reduce or inhibit the protein degradation. At the antepenultimate position from the C terminus, residues with no side chain or acidic side chains can increase the stability, i.e. reduce or inhibit the protein degradation. The combinational substitution of only the last three residues of polypeptides can make the polypeptides more stable during heterologous expression in mycobacterial hosts.

摘要: Methods and apparatus for planning and dynamically updating sampling operations while drilling in a subterranean formation are described. An example method of planning a sampling while drilling operation for a subterranean formation includes identifying a plurality of processes and related parameters, the processes including drilling and sampling processes and the related parameters including drilling and sampling parameters. The example method also involves processing the parameters for each of the processes via a simulation engine to generate predictions associated with sampling the formation, the simulation engine including at least one of a wellbore hydraulics simulator, a mudcake simulator, a formation flow simulator, or a tool response simulator. The example method also involves ranking the predictions associated with sampling the formation based on at least one of a sample fluid quality, a sampling process duration, a sampling process efficiency or a cost of sampling, and planning the sampling operation based on the ranked predictions.

摘要: The present invention relates to a subscriber station security-related parameter negotiation method in a wireless portable Internet system. The subscriber station security-related parameter negotiation method includes security-related parameters in transmitting/receiving basic capability negotiation request messages and basic capability negotiation response messages such that the subscriber station and the base station negotiate the subscriber station security-related parameters. The security-related parameters include an authorization policy support subfield used to negotiate an authorization policy between the subscriber station and the base station, and message authentication code mode subfields used to negotiate a message authentication code mode. The base station can inform the subscriber station that authentication or message authentication is not performed and is omitted according to a service provider policy by writing it on the authorization policy support subfield or message authentication code mode subfield. In addition, the subscriber station and the base station can select an authorization policy formed with at least one combination through the authorization policy support subfield of the security negotiation parameters. According to the present invention, the service provider of the wireless portable Internet system can more efficiently and flexibly manage the system by providing a scheme for omitting an authentication function and a message authentication function as well as for supporting various authorization policies and message authentication functions.

摘要: Methods of and apparatus to perform a drawdown of a formation fluid in a downhole environment are disclosed. An example method includes contacting a borehole wall with a fluid communication device of a formation testing tool and performing a first type of drawdown to draw fluid into the fluid communication device. The method also includes detecting a breach of a mudcake on the borehole wall during performance of the first type of drawdown and performing a second type of drawdown to draw fluid into the sample probe in response to detecting the breach of the mudcake. The second type of drawdown is different than the first type of drawdown. Furthermore, the example method includes confirming the breach of the mudcake on the borehole wall during performance of the second type of drawdown.
