HERV-K human endogenous retroviruses show up-regulated expression in tumors. In particular, splicing events in the env region generate a series of transcripts which utilise the +2 reading frame, relative to the env reading frame. The proteins show activity typical of transcriptional regulators, and they also have oncogenic potential. Two related proteins, PCAP2 and PCAP3, are strongly associated with breast cancer and prostate cancer, respectively. PCAP4 stimulates cell division. These proteins can be used in cancer diagnosis and therapy, and are also drug target e.g. for adjuvant therapy. The identification of these splice products is remarkable because full sequence information has been available for HERV-K viruses since 1986.

A specific member of the HERV-K family located in chromosome 22 at 20.428 megabases (22q11.2) has been found to be preferentially and significantly up-regulated in prostate tumors. The invention provides methods for diagnosing prostate cancer, comprising the step of detecting in a patient sample the presence or absence of an expression product of the virus. The virus has five features not seen in other HERV-K members: (1) its own specific nucleotide sequence, and consequently amino acid sequences; (2) tandem 5′ LTRs; (3) a fragmented 3′ LTR; (4) an env gene interrupted by an alu insertion; and (5) unique gag sequences.

DNA sequences encoding human platelet-derived growth factor receptors (hPDGF-R), and expression constructs comprising sequences which encode a receptor that can be secreted or incorporated into the membrane of a mammalian cell. Peptide fragments with functions equivalent to the wild-type receptor, conferring a PDGF-sensitive mitogenic response on cells lacking the receptor are provided. The constructs can be used for enhancing PDGF response of cells, determining the regions involved in transducing the signal in response to PDGF binding, providing mutated analogs and evaluating drugs for their physiologic activity. Soluble fragments comprising PDGF receptor sequences are also provided, including important intracellular kinase insert sequences which interact with intracellular proteins.

KIAA0175 inhibitors include anti-sense molecules, ribozymes, antibodies and antibody fragments, proteins and polypeptides as well as small molecules. KIAA0175 inhibitors find use in compositions and methods for decreasing KIAA0175, p53 and/or p21 gene expression as well as methods for increasing the chemo and/or radiosensitivity of mammalian cells, including tumor cells, methods for decreasing the side effects of cancer therapy and methods for treating neoplastic diseases.

During the testing of circuit boards with Automatic Test Equipment in some circuit configurations the performance of a component may not be as expected even though it is sound due to the influence of surrounding components. In such a situation guards may be placed, that is further signals via probes contacting nodes other than those to which the component is connected, so that such disruption may be minimized. A method for defining guards during the operation of automatic test equipment so that a selected component of a circuit may be tested includes establishing a measurement of the selected component in a known to be good circuit of the type to be tested. A circuit node associated with the selected component is identified and a guard is applied to the node. A second measurement is established with the selected guard in place. If the guard results in an improvement, it is retained during subsequent automatic testing.

Additionally, an optical scanner is positioned adjacent the transparent scanning surface. The optical scanner detects contrasting markings on the sheets of media through the transparent scanning surface. Vibrating elements are connected to the transparent scanning surface.
