This invention relates to immunoglobulin molecules comprising light chain (VL) chimeric variable domains, heavy chain (VH) chimeric variable domains, e.g., scFv antibodies that are expressed at high levels within a host cell, preferably within particular cellular compartments such as, e.g., cytosol or apoplast. The VL, VH and scFv antibody molecules comprise framework scaffolds of particularly preferred framework regions. This invention also relates to nucleic acid molecules encoding the immunoglobulin molecules of this invention, vectors expressing the immunoglobulin molecules, hosts transformed with the nucleic acid molecules and vectors, and methods of using the immunoglobulin molecules. Also described are immunoglobulin libraries as well as host cells, including transgenic plants, expressing the VL, VH or scFv antibody molecules of this invention.

The present invention is drawn to a fusion protein containing at least one binding domain that specifically recognizes an eptitope of a plant pathogen and at least one additional domain made from a protein or peptide sequence which is toxic to the pathogen or detrimental the replication, transmission or life cycle of the pathogen. The present invention is further drawn to a pathogenocide made from the binding domain, a cellular targeting sequence and/or membrane localization sequence that leads to membrane anchoring. The present invention is further drawn to nucleotide sequences encoding fusion proteins and pathogenocides and to vectors containing the nucleotide sequences; as well as methods of making the fusion proteins and pathogencides and methods of making pathogen resistant plants, plant cells, or plant tissues with the fusion proteins and pathogenocides.

The invention provides a method for the generation of a monoclonal plant cell line from a heterologous population of plant cells, comprising the following steps: (a) provision of a heterologous population of plant cells; (b) preparation of protoplasts from said heterologous population of plant cells; (c) separation of single protoplasts by subjecting the preparation of protoplasts to flow cytometric sorting; (d) regeneration of a separated single transformed protoplast until the formation of a microcolony by co-cultivation in the presence of feeder cell material; (e) removal of the microcolony from the feeder cell material and cultivation of the microcolony until the formation of a monoclonal plant cell line.

The present invention relates to fusion proteins comprising anti-Oomycotic proteins or peptides linked to an antibody or fragment thereof specifically recognising an epitope of an Oomycota. The invention is also directed to polynucleotides coding for the fusion proteins. The embodiments of the present invention are particularly useful for the protection of plants against Oomycota. The invention therefore comprises transgenic plants expressing the fusion proteins of the present invention.

The present invention relates to a method for identifying compounds that affect a transport of a receptor of interest through a specific membrane trafficking pathway mediated by said receptor within the context of a generic membrane trafficking pathway, which generic pathway is not mediated by said receptor, characterised by the following steps: d) monitoring the transport of the receptor of interest in a cell in the presence of the compound, wherein the receptor is labelled with a first marker, e) monitoring the transport of a second marker through the generic membrane trafficking pathway in a cell in the presence of the compound, f) comparing the results obtained from steps a) and b) and thereby identifying a compound affecting the specific membrane trafficking pathway. Furthermore it relates to the generation of a compound database using the method according to the invention as well as to the compound database itself.
