The present invention relates to a method for controlling carbon source concentration in the aerobic cultivation of a microorganism. The substrate carbon source remains at a low level in a cultivation vessel during the culture feeding in aerobic fed-batch, continuous or cell-recycling continuous cultures. This is accomplished by monitoring the increase in pH or dissolved oxygen content in the culture medium and adding the feed solution intermittently into a cultivation vessel at a calculated feed rate using a feed control device controlled by a computer.The present invention further relates to a process for producing L-lysine by fermentation having the advantages over the prior methods, those being improved productivity, higher concentrations of accumulated product, and increased yields of L-lysine.

An apparatus and method for defoaming aerobic fermentation cultures is provided, where the amount of liquid culture medium used is greater than that in conventional aerobic cultures (70% or more of the total capacity of the culture tank) without lowering the productivity of the intended, desired product. In the present apparatus and method, a device for separating vapor from liquid of a foam is in fluid communication with the fermentation tank, a device for condensing residual liquid of the vapor received from the separating device is in fluid communication with the separating device, and a sensor for detecting foams is provided in a location along the fluid communication pathway. In the defoaming apparatus, an optional defoaming device is provided, which may be based on either a rotary body rotating at a high speed by use of an electric motor which beats the foams, or on a centrifugal atomizer. The defoaming device may be provided between the vapor-liquid separating device and the condensing device.

The flows in the orifices and the flows directly below the orifices are configured to be substantially liquid film flows. The directions of jet flows (30), (31) from the respective orifices (12) are not necessarily matched to the central axis directions of the orifices and are not necessarily intersected with each other at a downstream position thereof. The sprays are caused to converge by the Coanda effect acting on a plurality of sprays after jet flows from the orifices (12) become sprays at a downstream position farther than a break-up length (a). The convergence of the sprays is continued until the Coanda effect is substantially lost.

The present invention is a method for aerobically cultivating yeast or bacteria in a culture medium of fed-batch, continuous or cell-recycling continuous cultures, wherein the carbon source concentration in the culture medium is maintained at a constant low level of under g/l. The carbon source concentration is maintained by measuring the carbon consumption of a culture of the yeast or bacteria in a preliminary experiment. The rate is determined between the time the culture is started and a time when the carbon source is exhausted. A feeding time is then determined wherein the activity of the yeast or bacteria in the presence of the carbon source does not change and a volume of the carbon source to be used in a first feeding (So) is set as So=.nu..times.T. Then, in a main culture, a first feeding of a volume of the carbon source (So) is added for the time (T), and the exhaustion of the carbon source is detected as an increase in pH or an increase in concentration of oxygen dissolved in the culture medium. A second feeding of a volume of the carbon source is then added for the time (T), and the feeding rate is determined based on a period (.tau.) before which the second feeding is added as follows:if 10 min..gtoreq..tau., the feeding rate of the feed solution is set at 1.1.times..nu.;if 30 min..gtoreq..tau.>10 min., the feeding rate of the feed solution is set at .nu.;if 60 min..gtoreq..tau.>30 min., the feeding rate is set at 0.9.times..nu.; orif 120 min..gtoreq..tau.>60 min., the feeding rate is set at 0.8.times..nu..
