    摘要: A dehydrated composition is provided that includes freeze-dried platelets. The platelets are loaded with trehalose which preserves biological properties during freeze-drying and rehydration. The trehalose loading is conducted at a temperature of from greater than about 25null C. to less than about 40null C., most preferably at 37null C., with the loading solution having trehalose in an amount from about 10 mM to about 50 mM. These freeze-dried platelets are substantially shelf-stable and are rehydratable so as to have a normal response to an agonist, for example, thrombin, with virtually all of the platelets participating in clot formation within about three minutes at 37null C.

The method comprises allowing output pressure of the pump to decrease over time t1, increasing output pressure of the pump over time t2, comparing achieved pump output pressure to a predetermined pressure at about the end of interval f, and at least one of (i) adjusting t1 and t2 if necessary so the predetermined pressure is approximated by the output pressure at the end of the next interval f, and (ii) adjusting a rate of change of the output pressure during at least one of t1 and t2 if necessary so the predetermined pressure is approximated by the output pressure at the end of the next interval f.

    摘要: A method for chemical fixation of tissues by exposing the tissue to a chemical fixative agent, under oxidative conditions. The chemical fixative agents useable in this method include aldehydes (e.g., formaldehyde, glutaraldehyde, dialdehyde starch), isocyanates (e.g., hexamethylene diisocyanate) and certain polyepoxy compounds (e.g., DENACOL). The oxidative conditions may be provided by heating of a chemical fixative solution that contains the crosslinking agent, in the presence of room air or oxygen. Alternatively, the oxidative conditions may be provided by adding one or more oxidizing chemicals (e.g., hydrogen peroxide or other peroxides, sodium periodate or other periodates, diisocyanates, halogens, n-bromosuccinimide or other halogenated compounds, permanganates, ozone, chromic acid, sulfuryl chloride, sulfoxides, selenoxides, etc.) to the chemical fixative solution. Alternatively, the oxidative conditions may be provided by irradiation (e.g., alpha, beta, ultraviolet, electron beam, gamma rays) of the fixative solution in the presence of room air or oxygen.

    摘要: Cell membrane maintenance of red blood cells and platelet concentrates is improved by the addition of 1 mM-10 mM L-carnitine and derivatives. This improvement allows extension of the period of viability of packed red blood cells and platelet concentrations beyond current periods. Additionally, the materials so treated exhibit extended circulation half life upon transfusion to a patient. Improvements in membrane maintenance achieved by this method permit irradiation of sealed containers of blood products so as to substantially sterilize and destroy leukocytes in the same.
