A method is provided for preparing high purity levoglucosan from lignocellulosic pyrolysis oils derived from wood or waste newsprint. The method includes reducing wood or newsprint to fine particle sizes, treating the particles with a hot mineral acid for a predetermined period of time, and filtering off and drying resulting solid wood or newsprint material; pyrolyzing the dried solid wood or newsprint material at temperatures between about 350.degree. and 375.degree. C. to produce pyrolysis oils; treating the oils to liquid-liquid extraction with methyl isobutyl ketone to remove heavy tar materials from the oils, and to provide an aqueous fraction mixture of the oils containing primarily levoglucosan; treating the aqueous fraction mixtures with a basic metal salt in an amount sufficient to elevate pH values to a range of about 12 to about 12.5 and adding an amount of the salt in excess of the amount needed to obtain the pH range to remove colored materials of impurities from the oil and form a slurry, and freeze-drying the resulting slurry to produce a dry solid residue; and extracting the levoglucosan from the residue using ethyl acetate solvent to produce a purified crystalline levoglucosan.

Process of preparing 1,6-.beta.-D-anhydroglucopyranose (levoglucosan) in high purity is described, where the starting material with a water-content, adjusted to max. 20%, is pyrolyzed, the volatile components are condensed and this distillate is neutralized. After filtration and optional treatment with an adsorbent, the mixture is chromatographically separated over an ion-exchange resin with water as eluent. Levoglucosan containing fractions are concentrated and crystalline levoglucosan is obtained by an evaporation - and/or cooling crystallization from a supersaturated water solution.

The invention relates to an enzyme, exo-.alpha.-1,4-glucan lyase, capable of successively cleaving the terminal .alpha.-1,4-D-glucosidic bonds from the non-reducing ends of an .alpha.-1,4-glucan. The enzyme is selected from the group consisting of .alpha.-1,4-glucan lyase isolated from an alga and functional derivatives and analogs derived from a nucleotide sequence related to an alga .alpha.-1,4-glucan lyase. Further, a method of enzymatically cleaving the terminal .alpha.-1,4-D-glucosidic bonds from the non-reducing ends of an .alpha.-1,4-glucan, and the degradation product 1,5-anhydrofructose for use as an oxygen radical scavenger or anti-oxidant and/or as a sugar substitute, are disclosed. Additionally, a method of producing an enzyme capable of successively cleaving the terminal .alpha.-1,4-glucosidic bonds from the non-reducing ends of an .alpha.-1,4-glucan; an antibody which binds to the amino acid sequence of the enzyme, and a DNA or RNA probe which recognizes a nucleotide sequence coding for the enzyme, are described.
