摘要： A nanobody or an antigen-binding fragment thereof against a specific region of a factor XII (FXII) is provided. The nanobody or the antigen-binding fragment thereof binds to FXII through a binding epitope for FXII to block the activation of FXII, and the binding epitope for FXII includes a conformational epitope. A neutralizing antibody for FXII screened by phage display is provided, and it has been found that the neutralizing antibody can treat thrombosis, myocardial ischemia-reperfusion injury (IRI), and vasculitis in mouse arterial thrombosis, rat extracorporeal membrane oxygenation (ECMO), mouse IRI, and vasculitis models.

摘要： A method of isolating an antibody is disclosed. The method comprises contacting a hydrophobic chelator, a non-ionic detergent and metal ions so as to generate an aggregate comprising the hydrophobic chelator, the detergent and the metal ions; and contacting the aggregate with a medium comprising the antibody under conditions that allow partitioning of the antibody into the aggregate. Kits for isolating the antibody are also disclosed.

摘要： The present disclosure provides antibodies that bind to the catalytic domain of Factor XI (FXI) and methods of using the same. According to certain embodiments, the antibodies are antagonist antibodies that inhibit blood clot formation via the intrinsic pathway without affecting hemostasis, as shown by their effect on prolonging aPTT without affecting PT. As such, these antagonist antibodies may be used to treat blood clotting diseases or disorders or treatment regimens that have clot formation as a risk factor, such as, but not limited to atrial fibrillation. In certain embodiments, the disclosure includes antibodies that bind FXI and mediate clot formation or thrombogenesis. The antibodies of the disclosure may be fully human, non-naturally occurring antibodies.

摘要： The present invention provides efficient methods based on alteration of the protein A-binding ability, for producing or purifying multispecific antibodies having the activity of binding to two or more types of antigens to high purity through a protein A-based purification step alone. The methods of the present invention for producing or purifying multispecific antibodies which feature altering amino acid residues of antibody heavy chain constant region and/or variable region. Multispecific antibodies with an altered protein A-binding ability, which exhibit plasma retention comparable or longer than that of human IgG1, can be efficiently prepared in high purity by introducing amino acid alterations of the present invention into antibodies.
