The pump radiation source emits a beam (900) of pump radiation having a profile comprising no pump radiation in a central region of the beam and excites the high harmonic generation medium so as to generate high harmonic radiation (920). The pump radiation (950) and the generated high harmonic radiation are spatially separated beyond the focal plane of the beam of pump radiation. The spatial filter is located beyond a focal plane of the beam of pump radiation, and blocks the pump radiation. Also disclosed is a method of generating high harmonic measurement radiation optimized for filtration of pump radiation therefrom.

The multi-channel fluorescence measuring optical system, which irradiates light onto a plurality of sample channels and detecting fluorescence radiated from samples, includes: a light source (60a-d); an integrator (70) for giving the light irradiated from the light source a uniform intensity distribution; a sample holder (80) having a plurality of sample channels on which the samples (m) are mounted, wherein the samples are exited by the light emitted from the integrator; and a beam splitter (75) between the integrator and the sample holder for dividing the incident light in a predetermined ratio. Since the light intensities of fluorescence images are detected using optical fiber bundles (85a-d) and photodiodes (89a-d), the manufacturing cost can be greatly reduced, and the optical system can be miniaturized.

Erfindungsgemäss wird darum eine ringförmige Lichtquelle vorgeschlagen, die im Bereich der optischen Achse eine verringerte Leuchtdichte aufweist, was ermöglicht, eine praktisch homogene Beleuchtungsstärke oder Flächenhelligkeit zu erzeugen. Als bevorzugte ringförmige Lichtquelle wird ein faseroptisches Ringlicht verwendet.

A flow cytometry apparatus for determining one or more characteristics of particles or the like flowing in a liquid stream includes a nozzle for generating a liquid flow stream for moving particles therethrough substantially one at a time. An arc lamp provides a beam of incoherent light to illuminate the particles moving in the flow stream. A prism in the light beam modifies non-uniform incoherent light, as provided by said arc lamp, to a more uniform beam of illumination which is directed to the flowing particles. A detector is included for detecting light with respect to each moving particle and for associating the detected light with one or more characteristics of the particles.

To each light source there corresponds a beam shaper comprising a light collector, a homogenizing means for light from the light source, and an imaging means for imaging an output of the homogenizing means into an area to be illuminated. The homogenizing means in embodiments is a rod, into which light from the light collector is directed. The end of the rod opposite the light collector is imaged by the imaging means into the area to be illuminated.
