Disclosed is an in vitro method for assessing the presence of gingivitis in a human subject. The method is based on the insight to determine a selection of three biomarker proteins. Accordingly, in a saliva sample of the subject the concentrations are measured of the proteins Hepatocyte growth factor (HGF) and Interleukin-1β (IL1β), and at least one of C reactive protein (CRP) and Haemoglobin. Based on the concentrations as measured, a value is determined reflecting the joint concentrations for said proteins. This value is compared with a threshold value reflecting in the same manner the joint concentrations associated with the absence of gingivitis or periodontitis. The comparison allows to assessing whether the testing value is indicative of the presence of gingivitis in said subject Thereby, typically, a testing value reflecting a joint concentration above the joint concentration reflected by the threshold, is indicative of the presence of gingivitis.

Methods for diagnosing the status of periodontitis disease includes selecting a set of protein biomarkers including one or more biomarkers which have been shown to vary in abundance at particular stages of periodontitis. The set of protein biomarkers may be identified and quantified in expression in an acquired gingival crevicular fluid (GCF) or saliva oral fluid sample in order to distinguish between different states of periodontitis. Methods of diagnosing the status of periodontitis oral disease at varying levels of severity, e.g. gingivitis, mild periodontitis, or severe periodontitis, may include selecting a set of protein biomarkers which are capable distinguishing between different stages of periodontitis.

A method of assessing a gum condition of an individual is provided, which comprising the steps: (a) tooth brushing within an oral cavity of the subject individual; (b) post-brushing rinsing the brushed oral cavity with a post-brushing rinse solution comprising volume from 1ml to 50 ml, for a predetermined post-brushing rinsing time, wherein the predetermined post-rinsing time is from 1second to 1 minute; (c) collecting expectorated post-brushing rinse solution from the subject individual; (d) measuring hemoglobin quantity contained in the collected post-brushing rinse solution; (e) comparing the measured hemoglobin quantity against a known standard of gum conditions corresponding to hemoglobin quantities to assess the gum condition of the subject individual from the compared hemoglobin quantities.
