The present invention provides a method for producing a myocardial sheet using a group of cells derived from embryonic stem cells. This method is characterized by mixing Flk/KDR positive cells, cardiomyocytes, endothelial cells, and mural cells, all derived from embryonic stem cells, and culturing the mixed cells. Furthermore, the myocardial sheet can be used as a therapeutic agent for heart diseases since VEGF is released from the sheet.

Methods and reagents are described for myocardial regenerative therapy using cardiomyocytes that are cultured ex vivo to have a controlled orientation of beating. This is achieved by growing the cardiomyocytes on a culture support that is coated with a polymerizable liquid crystal which is oriented so as to be in a liquid crystal phase state prior to curing with ionizing radiation or ultraviolet rays.

Provided are a cell sheet and a three-dimensional structure thereof that include at least mesenchymal stem cells and myoblasts isolated from a cell culture support and that are to be applied to heart disease. Use of the cell sheet and the three-dimensional structure thereof including mesenchymal stem cells and myoblasts can notably improve cardiac functions compared with a conventional technology, i.e., use of cell sheets composed of myoblasts only or mesenchymal stem cells only. Furthermore, the cell sheet itself has high strength, and the transplantation procedure is also improved.

A method for manufacturing a multilayered cell sheet characterized in fabricating a vascular bed that constructs a vascular network extending to the surface from a channel for perfusing a medium, the channel being embedded in a gel; and layering a cell sheet onto the vascular bed to construct a vascular network in the cell sheet. This manufacturing method makes it possible to construct a vascular network in the cell sheet and to fabricate a thick multilayered cell sheet in a simple manner by layering cell sheets. Such a thick multilayered cell sheet is useful as an in-vivo tissue substitute in regenerative medicine involving a variety of tissues.

This invention provides a cell pattern recovery tool comprising a base material layer having a surface subjected to easy adhesion treatment, a temperature responsive polymer layer that is provided on the base material layer and has a surface subjected to silane treatment, and a cell adhesion inhibiting material layer provided on the temperature responsive polymer layer. According to the present invention, a cell pattern can be rapidly recovered while maintaining the cell pattern stably and reliably under minimally invasive conditions for the cells.

A substrate surface having a cell non-adhesive region on the periphery of a cell adhesive region is used, and (1) the cells are allowed to adhere to the cell adhesive region of the substrate surface in a differentiation-inducing medium; (2) the cells are subsequently allowed to grow on the cell adhesive surface to reach confluence; (3) the culture is further continued to three-dimensionally stratify the cells from the boundary with the cell non-adhesive region surrounding the cell adhesive region; and (4) a plurality of the stratified portions formed in the cell adhesive region is finally bound to one another to produce a cell agglomerate spreading over the entire cell adhesive region with a certain thickness.
