摘要: The present invention relates to methods for culturing and isolating cells such as multipotent stem cells or multipotent progenitor cells. The methods of the invention comprise using the human CD105 polypeptide as a cell culture substrate. The cells obtained according to the methods of the invention are capable of forming colony forming unit-fibroblasts (CFU-Fs). Cell lines comprising such cells are useful for the treatment of medical conditions, such as acute respiratory distress syndrome (ARDS).

摘要: In certain aspects, the present invention provides methods for inducing a stable gene modification of a target nucleic acid via homologous recombination in a primary cell, such as a primary blood cell and/or a primary mesenchymal cell. In certain other aspects, the present invention provides methods for enriching a population of genetically modified primary cells having targeted integration at a target nucleic acid. The methods of the present invention rely on the introduction of a DNA nuclease such as a Cas polypeptide and a homologous donor adeno-associated viral (AAV) vector into the primary cell to mediate targeted integration of the target nucleic acid. Also provided herein are methods for preventing or treating a disease in a subject in need thereof by administering to the subject any of the genetically modified primary cells or pharmaceutical compositions described herein to prevent the disease or ameliorate one or more symptoms of the disease.

摘要: Stromal stem cells are prospectively isolated from human bone marrow then expanded into clonal populations and cultured and used, the isolation being on the basis of expression of a cell surface marker, wherein the cell surface marker binds an antibody and wherein said antibody cross reacts with a cell surface marker found on mouse stromal stem cells or rat stromal stem cells, and optionally also on a cell of at least one other mammalian species selected from mouse, rat, horse, rabbit and pig cells. Useful stromal stem cell populations are positive for SDC2.

摘要: The present invention relates to a medium composition for reinforcing the efficacy of stem cells, including ethionamide, a method of reinforcing the efficacy of stem cells, including culturing stem cells in the medium composition, a method of preparing stem cells with reinforced efficacy, stem cells prepared by the above-mentioned method, and a use thereof. According to the present invention, the anti-inflammatory effect of mesenchymal stem cells and expression levels of paracrine factors may be effectively improved by a simple method of treating mesenchymal stem cells with ethionamide, and the stem cells obtained by the above method may be effectively used for preventing or treating an inflammatory disease or a degenerative brain disease.
