摘要: Described herein are reprogrammed cells, and methods for cell dedifferentiation, transformation and eukaryotic cell reprogramming. Also described are cells, cell lines, and tissues that can be transplanted in a patient after steps of in vitro dedifferentiation and in vitro reprogramming. In particular embodiments the cells are Stem-Like Cells (SLCs), including Neural Stem-Like Cells (NSLCs), Cardiac Stem-Like Cells (CSLC), Hematopoietic Stem-Like Cells (HSLC), Pancreatic Progenitor-Like Cells, and Mesendoderm-like Cells. Also described are methods for generating these cells from human somatic cells and other types of cells. Also provided are compositions and methods of using of the cells so generated in human therapy and in other areas.

摘要: Microfluidic “organ-on-a-chip” devices have been developed with the aim to replicate human tissues in vitro. However, there is no option to quantitatively monitor biological processes that take place within the chip, over time. Destructive methods in order to analyze, tissue formation, gene expression, protein secretion etc. require the harvest of the “tissue” at a certain time point. Described herein are methods and compositions for non-destructive molecular imaging methods and systems in order to quantitatively monitor specific biological processes, over time, within the chip, without the need to harvest.

According to the present invention, there is provided a method that is a quality management method for a specific cell, where the method includes evaluating a correlation relationship between an expression pattern of a gene group obtained by measuring an expression state of a related gene group that is characteristic of the specific cell and an expression pattern that serves as a reference and determining the cell to be usable in a case where the correlation relationship is equal to or higher than a certain level.

摘要: The present invention relates to a composition containing primed, function-reinforced stem cells for preventing, alleviating or treating atopic dermatitis. The stem cells primed by treatment with TNF-α, IFN-γ, and IFN-α or with TNF-α, IFN-γ, IFN-α, and vitamins according to the present invention have reinforced anti-inflammatory and immunomodulatory ability and thus exhibit excellent prophylactic or therapeutic effects on atopic dermatitis symptoms, compared to stem cells that are not treated with the above priming factors. Therefore, the function-reinforced stem cells of the present invention can be widely utilized in various fields of prevention, alleviation, or treatment of atopic dermatitis.

摘要: Forced expression of a handful of transcription factors (TFs) can induce conversions between cell identities; however, the extent to which TFs can alter cell identity has not been systematically assessed. Here, we assembled a human TFome, a comprehensive expression library of 1,578 human TF clones with full coverage of the major TF families. By systematically screening the human TFome, we identified 77 individual TFs that induce loss of human-induced-pluripotent-stem-cell (hiPSC) identity, suggesting a pervasive ability for TFs to alter cell identity. Using large-scale computational cell type classification trained on thousands of tissue expression profiles, we identified cell types generated by these TFs with high efficiency and speed, without additional selections or mechanical perturbations. TF expression in adult human tissues only correlated with some of the cell lineage generated, suggesting more complexity than observation studies can explain.

摘要: A method of making a 3D-printed biomaterial is described. The method includes depositing a hydrogel comprising cells and a cryoprotectant from a 3D printer onto a freezing plate to form a frozen hydrogel filament; depositing additional hydrogel comprising cells and a cryoprotectant from a 3D printer in contact with a previously placed frozen hydrogel filament to form a 3D-printed biomaterial comprising a plurality of frozen hydrogel filaments; and removing the 3D-printed biomaterial from the freezing plate. 3D-printed biomaterials made by the method are also described.
