    摘要: A method of sex sorting sperm is disclosed. The sperm may be stained with a DNA selective fluorescent dye, which fluoresces when excited, a dead quenching dye, which selectively quenches fluorescence emitted by the DNA selective fluorescent dye within the membrane of compromised sperm, and a split enhancing dye. The stained sperm may then sorted into one or more gender enriched subpopulations of viable sperm.

    摘要: The present invention addresses the problem of providing an agent for improving sperm-motility, in particular, an agent for improving forward-sperm-motility, for use on reduced-motility sperm in male infertility treatment. Prepared is a gaseous agent for improving sperm-motility consisting of a gas that contains hydrogen molecules in the amount of 1% (v/v) or higher, for example, a gas that contains hydrogen molecules in an amount of between 45 and 55% (v/v), or a liquid agent for improving sperm-motility consisting of a liquid such as physiological saline, a culture solution, or a buffer solution containing hydrogen molecules in the amount of 1% or higher of the saturation solubility thereof, for example, a liquid in which hydrogen molecules have been dissolved by bubbling. The gaseous agent for improving sperm-motility can improve the motility of sperm by contacting hydrogen gas and either semen or a dilution thereof with each other in a gaseous phase. The liquid agent for improving sperm-motility can improve the motility of sperm when used as a washing solution or diluent for semen.

    摘要: A sperm diluent of the present invention contains a chelating agent such as EDTA and/or, EGTA, which forms a complex with a calcium ion, in a base diluent. Further, the sperm diluent contains an immunosuppressive factor such as a steroid hormone and/or, a cytokine, which suppresses migration of leukocytes. By diluting frozen sperm with this sperm diluent followed by performing artificial insemination, death of the sperm before fertilization and phagocytosis of the sperm and embryos by leukocytes in the uterus can be suppressed, allowing enhancement of the conception rate and the implantation rate.

The present invention relates to novel compounds that can be added to the sperm cell sample to reduce the traumatic effects of physical stress during mild as well as extensive sperm cell processing, methods of using the compounds in standard sperm processing procedures, the end products made from these methods including sperm and embryos, as well as methods of using those end products in assisted reproductive biology techniques in animals.

    摘要: The present invention provides an aptamer or pool of aptamers (nucleic acid sequences) that bind(s) to a target molecule on the surface, accessible from the surface or inside of a mammalian sperm cell and a method for producing the aptamers. The method comprises contacting a collection of different nucleic acid molecules with the target molecule under conditions favorable for binding between at least one of the nucleic acid molecules and the target molecule, to form at least one complex comprising the nucleic acid molecule bound to the target molecule, wherein each of the nucleic acid molecules comprises at least one segment of randomized nucleotide sequences. The complexes are then separated from the unbound nucleic acid molecules and unbound target molecules, and the bound nucleic acid molecule is recovered from the separated complex. The aptamers are used to separate sperm cells based upon sperm cell qualities, such as whether the cells contain X chromosomes or Y chromosomes.
