An alternating current input to direct current output power supply providing a regulated constant voltage output or constant current output and incorporating a switching-mode regulating element. Regulation is primarily obtained by means of a bi-directional switching element operated at a relatively low voltage, and the alternating current voltage output from the switching element and an associated power transformer is applied to a rectifying voltage multiplier to produce a high voltage output. The voltage multiplier is subject to receiving an unbalanced potential, thereby imposing a dc off-set potential onto the switching element to cause instability in the potential at the power supply output. A correcting circuit is provided to eliminate the effect of the dc off-set potential on the switching element.

A chromatographic monitor includes an array of flow cells with individual light sensors that are collectively an array of photodiodes. The output from the photodiodes are multiplexed. To prevent losing information, the photodiodes are each connected to a different one of a plurality of inputs to the multiplexer through a corresponding one of a plurality of circuits that stores energy during the time the one inlet is not connected through the multiplexer to the signal processing circuitry that forms a part of an absorbance monitor. Preferably, the energy storing circuit is a non-switching circuit with low bandwidth and a flat-topped response to an impulse. This improves the signal to noise ratio. A one pole low pass filter with a (1-1/e) Dirac pulse fall time and equal to the multiplex entire cycle repeat time can perform this function and a one pole low pass filter provides satisfactory results. Still better results can be obtained from a three pole, one or two percent overshoot filter with combined minimum frequency bandwidth and fast rise time. A rise time equal to ½ the multiplex entire cycle.

To determine the total organic carbon content of a flowing liquid in a continuous process, water and oxidizing agents enter lower ports in a continuous column and flow upwardly along a helical path with air gaps as they are mixed. The helical reaction path is formed by a helical glass cylinder between an ultraviolet lamp and outer wall of a continuous glass tube to cause rapid motion of the liquid. An ultraviolet lamp extends downwardly through the center of the column so that the outer confining surface of the helical path is the outer surface of the ultraviolet lamp, causing the liquid to move rapidly in intimate contact with the ultraviolet lamp to receive intense radiation. An air gap permits escape of air and reduces bubble size to reduce plug flow.

An apparatus for supercritical fluid extraction incorporates a removable extraction cartridge which in operation has insignificant pressure difference between its inside and outside walls. Because of the low pressure difference, the extraction cartridge need not have the strength to withstand significant pressure and can be made out of molded plastic for disposable use as well as stainless steel and/or machined plastic for reusability. The extraction cartridge can be removed and opened for sample access without the use of tools. The outside of the cartridge can be purged after it is installed in a heated high pressure vessel to remove contamination from its exterior. In one embodiment, the extractor includes a fraction collector for extractants, an automatic sample changer and an automatic cartridge transfer mechanism which provide completely automated extractions.

To reduce noise in an absorbance monitor in which the light source is a deuterium lamp, an aspheric condensing mirror focuses light from an approximately 0.6 mm-high by 2 mm-wide part of the central bright spot in the deuterium lamp through a slit in an aperture plate which blocks all other light and an aspheric monochromator mirror focuses the light onto a plane reflective diffraction grating, which: (1) is mounted for orbiting about an axis parallel to the longitudinal axis of the slit; and (2) adjusted so that, as it orbits it reflects a selected wavelength of light to a beam splitter. Each beam is focused upon and transmitted through a different one of two equal oval apertures optically at least as large as the height of the image of the aperture near the lamp. The beams are converted to an electrical signal by silicon diode photosensors, thus using only light: (1) focused in both transverse planes and originating from the same spot on the central bright spot; and (2) with the same relative geometric weighting for light propagating in various directions from various subparts of the central bright spot. To reduce noise originating from flow-related refraction effects in the flow cell, the flow cell has reflective walls and a large collecting mirror focuses light entry in a large cone from the flow cell into a measuring photocell. This large cone has a wider angle than the cone of light entering the flow cell.

The bottoms of the wells are covered with clamped-on semipermeable membrane assemblies to permit the flow of buffer ions but not the migration of separated sample. Electrical continuity for the electrophoretic migration taking place in capillary tubes is provided through the electrolyte in wells, the assembled semipermeable membranes, electrolyte buffer residing the carrier, the electrode and a conductor leading to electrical ground. Separated zones are detected, electrophoretically eluted or electrosmotically discharged from the capillary tube into the electrolyte in well where they are trapped by the semipermeable membrane in assembly.

To supply a programmed gradient to a high pressure pump for introduction into a column, a mixer, degasser and accumulator communicates with the high pressure pump to supply fluid thereto, having a volume no more than 150 times the chamber volume of the high pressure pump. The mixer, degasser and accumulator includes temperature sensors which sense when the mixer, degasser and accumulator has been emptied to a level where it cannot respond to a demand for fluid from said high pressure pump and provides a signal to a low pressure pump which responds by cycling to again fill the mixer, degasser and accumulator. Upon receiving a demand signal, the low pressure pump fills by drawing fluid from a plurality of fluid sources to compose the gradient being used at that time, with the pump slowing during valve opening and closing so as to avoid cavitation.

To detect a ground fault, the alternating current leakage current to common AC electrical ground is sensed across a resistor and applied to one input of an analog gate. The analog gate is opened by the positive going voltage from the high-voltage secondary of the power supply transformers between 7 degrees from the crossover point and 103 degrees to pass only that portion of the potential representing the alternating current leakage current which is in phase with the potential on the secondary windings of the power supply transformer and thus provide a signal representing the real component of the AC impedance to ground. A reference preset level of impedance and the signal from the analog gate are compared to provide a signal whenever the real component of the AC leakage current exceeds the preset value.
