Methods for making biomaterials for use as a tissue sealant, kits containing precursors for forming the biomaterials, and the resulting biomaterials are described herein. The biomaterials are formed from a composition comprising at least a first and a second precursor molecule, wherein: i) the first precursor molecule is a poly(ethylene glycol) based polymer having x nucleophilic groups selected from the group consisting of thiol or amino groups, wherein x is greater than or equal to 2 ii) the second precursor molecule is of the general formula: A-[(C3H6O)n—(C2H4O)m—B]i wherein m and n are integers from 1 to 200 i is greater than 2 A is a branch point B is a conjugated unsaturated group The precursors are selected based on the desired properties of the biomaterial. Optionally, the biomaterials contain additives, such as thixotropic agents, radiopaque agents, or bioactive agents. In the preferred embodiment, the biomaterials are used to reduce, inhibit, or contain loss of a biological fluid or gas in a patient.

A method for encapsulating biologics within a hydrogel by using an aqueous solution of an isocyanate-functional hydrogel prepolymer which is mixed with an amount of biologics and an aqueous solution containing a dithiol crosslinking agent under physiological pH conditions. An additional bidentate crosslinking agent may be included. The product of such method may be a bioreactor or an assay device having a plurality or different biologics encapsulated at predetermined locations in a substrate.

A method for the preparation of highly elastic foams of reduced compression hardness and having urethane groups is disclosed, in which the reaction of the polyurethane-forming materials is carried in the presence of 0.01 to 5% by weight, based on the polyol, of a compound, which has at least one polyoxyalkylene group and at least one anionic group in the molecule. The use of fluorocarbons to decrease the compression hardness can reduced appreciably by the use of this compound.

Process for the production and purification of SH-terminated prepolymers by reaction of polyhydroxy compounds with thioalkane carboxylic acids, wherein one or more adducts of ethylene oxide and/or propylene oxide with triols or tetraols corresponding to the following general formula ##STR1## in which m=1, 2 or 3,n=0, 1 or 2 andp=0 or 1,and having a molecular weight of from 400 to 10,000 are reacted with one or more thioalkane carboxylic acids corresponding to the following general formulaHS--R--COOH (II)in which R is a straight-chain or branched C.sub.1 -C.sub.8 alkylene radical, in such a molar ratio that, for every mole of thioalkane carboxylic acid (II), there is an equivalent of the OH groups in compound (I), a neutralizing agent is added to the reaction mixture in a quantity sufficient to neutralize the unreacted quantity of thioalkane carboxylic acid (II), one or more compounds which bind the water of neutralization and a precipitant in excess are added at the same time as or immediately after the neutralizing agent, the reaction mixture is freed in known manner from the precipitated solids and, if desired, the prepolymer is isolated.
