摘要:
The present invention includes devices, systems, and methods for assaying cells using cell-substrate impedance monitoring. In one aspect, the invention provides cell-substrate impedance monitoring devices that comprise electrode arrays on a nonconducting substrate, in which each of the arrays has an approximately uniform electrode resistance across the entire array. In another aspect, the invention provides cell-substrate monitoring systems comprising one or more cell-substrate monitoring devices comprising multiple wells each having an electrode array, an impedance analyzer, a device station that connects arrays of individual wells to the impedance analyzer, and software for controlling the device station and impedance analyzer. In another aspect, the invention provides cellular assays that use impedance monitoring to detect changes in cell behavior or state. In some preferred aspects, the assays are designed to investigate the affects of compounds on cells, such as cytotoxicity assays. In other preferred aspects, the assays are designed to investigate the compounds that effect IgE-mediated responses of cells to antigens.

This application discloses a multivalent opsonophagocytosis assay that does not rely on counting of bacterial colonies to determine bacteria viability following opsonophagocytosis. Instead, the method uses a metabolic colorimetric indicator to determine if viable bacteria are present. Also disclosed are arrays that can be used to determine the viability of bacteria following opsonophagocytosis.

A method of using a microsample treatment apparatus and an apparatus for detecting chemotaxis of cells and separating chemotactic cells includes a number of wells that are connected to each other via a part having resistance to fluids. The wells are each provided with tubes for injecting/sucking a sample and, if necessary, tubes for relieving pressure changes at the injection/suction. The tubes have a space in common at the top ends thereof in which a liquid can be held.

Aspects of the present invention provide novel apparatus and methods for sampling microbial organisms present on surfaces. Preferred apparatus comprises: a reservoir suitable for providing microbial collection fluid; a sterilizable sample collection chamber; a sterilizable, integrated collection fluid delivery and collection fluid recovery member, suitable to deliver collection fluid to a target surface, and contemporaneously recover the delivered fluid from the surface; delivery means, in communication with both the reservoir and the integrated member, and operable to aseptically deliver collection fluid from the reservoir to the integrated member; and vacuum means, in communication with both the sample collection chamber and the integrated member, and operable to direct collection fluid, delivered and recovered by the integrated member, to the sample collection a method for rapid, high-throughput sampling of microbial organisms present on surfaces, comprising: delivering sample collection fluid to a target surface, and contemporaneously recovering the delivered fluid from the target surface by means of an integrated collection fluid delivery and collection fluid recovery member; and collecting the recovered sample collection fluid into a sample collection chamber in communication with the integrated member, whereby sample collection is, at least in part, achieved. The inventive methods and apparatus can be applied to both surface sampling and to atmospheric sampling.

The invention provides a method of determining a comparative expression profile in an individual by comparing the expression levels of a sample of molecules in a population of molecules in a specimen from the individual with a health-associated reference expression region of the sample of molecules, wherein expression levels within the health-associated reference expression region indicate a reference expression profile and wherein expression levels outside the health-associated reference expression region indicate a perturbed expression profile. The invention also provides methods of diagnosing a disease or a health state in an individual by comparing the expression level of a sample of molecules in a specimen from the individual with a health-associated reference expression region of the sample of molecules. The invention additionally provides a method of classifying a population by drug responsiveness.

The invention concerns a method for differentiating and counting cellular components present in a biological fluid sample comprising a primary cytological analysis step typically implemented by a flow cytometry equipment (1) to obtain a set of primary results enabling the set of cellular components of the sample to be differentiated and counted in different populations; and a complementary step of cytological analysis of a particular type of cellular components, based on an identified cellular peculiarity, to obtain complementary results enabling at least one cell population or subpopulation of the sample to be differentiated and counted for identification of said cellular peculiarity. The invention is applicable in particular to hematological analyses.
