摘要: A multiplexed enzyme assay method and substrate set for performing the method are disclosed. The method includes performing a plurality of enzyme reactions in the presence of a plurality of enzymes substrates, under conditions effective to convert an enzyme substrate to a corresponding product, where the product of each substrate and the substrate have different separation characteristics from each other and from the other substrates and their corresponding products. After performing the reactions, which may be carried out in separate or combined reactions, the substrates and products in said reactions are separated in a single separation medium. For each separated product and substrate, a separation characteristic effective to identify that product and substrate and a signal related to the amount of the product and substrate is detected. From this, one can determine the amount of substrate converted to the corresponding product in each of the reactions.

摘要: Novel methods and devices are provided involving at least one chamber, at least one capillary, and at least one reagent involved in a system providing for a detectable signal. As appropriate, the devices provide for measuring a sample, mixing the sample with reagents, defining a flow path, and reading the result. Of particular interest is the use of combinations of specific binding pair members which result in agglutination information, where the resulting agglutination particles may provide for changes in flow rate, light patterns of a flowing medium, or light absorption or scattering. A fabrication technique particularly suited for forming internal chambers in plastic devices is also described along with various control devices for use with the basic device.

摘要: An apparatus for diluting a sample with a diluent, comprising a fixed volume measuring chamber, a fixed volume receiving chamber and fluid receiving relationship to the measuring chamber, a gas vent in the receiving chamber, a stop flow junction between the measuring chamber and the receiving chamber, a sample application site in fluid donating relationship to the measuring chamber, wherein the vertical height difference between the sample application site and the stop flow junction is insufficient to provide a hydrostatic pressure capable of overcoming backpressure at the stop flow junction when sample is applied to the sample application site, and the diluent application site and fluid donating relationship to the measuring chamber. Sample and diluent are added sequentially to the apparatus of the invention without requiring any intervening operations. Various means are provided for restarting flow, which causes diluent to wash the sample into the receiving chamber, where the two can be mixed.

摘要: A method is disclosed for reducing the oxygen content of a medium in which are present cells, usually anaerobic microorganisms, to extend the time during which the cells remain viable. The method comprises having in fluid contact with the aqueous medium an effective amount of an oxidase and substrate for the oxidase. The oxidase and substrate for the oxidase in an aqueous medium can be in fluid, e.g., air, contact with a separately contained aqueous medium in which the cells are present. Alternatively, the cells can be present in the same aqueous medium as the oxidase and substrate for the oxidase. The aqueous medium containing the oxidase and the substrate for the oxidase can further contain a hydrogen peroxide scavenger in an effective amount. A composition in accordance with the invention comprises in a liquid medium viable cells, such as anaerobic microorganisms, an oxidase and its substrate in an amount sufficient to retain the viability of the cells, and a hydrogen peroxide scavenger in an amount sufficient to reduce the level of hydrogen peroxide generated by the oxidase. The method of the invention has particular application in transport media used for transporting microbiological specimens.

摘要: Enzyme channeling assay is provided employing a solid non-porous surface to which is bound a member of a specific binding pair and an enzyme. A complementary member of a specific binding pair is conjugated to second enzyme, so that the amount of second enzyme which becomes bound to said solid non-porous surface by the binding of a specific binding pair(s) is related to the amount of analyte in the liquid assay medium with which the solid surface is in contact. The first and second enzymes are related by the product of one being the substrate of the other. The turnover of the substrate formed by one of the enzymes results in a detectable product, which can be related to the amount of analyte in the medium.
