Abstract: The present technology provides for a microfluidic substrate configured to carry out PCR on a number of polynucleotide-containing samples in parallel. The substrate can be a single-layer substrate in a microfluidic cartridge. Also provided are a method of making a microfluidic cartridge comprising such a substrate. Still further disclosed are a microfluidic valve suitable for use in isolating a PCR chamber in a microfluidic substrate, and a method of making such a valve.

Abstract: Systems for processing a cellular sample are provided. Embodiments of the systems include a tray configured to receive a multi-microwell-array-flowcell cartridge, and a retrieval magnet assembly configured to apply a uniform magnetic force to flowcells of the cartridge when in an active position. Also provided are methods, which methods may include introducing a multi-microwell-array-flowcell cartridge into the system of the invention, loading a sample into the multi-microwell-array-flowcell cartridge, actuating the retrieval magnet assembly to the active position and applying the uniform magnetic force to the sample within the multi-microwell-array-flowcell cartridge to produce a processed sample, and collecting the processed sample from the multi-microwell-array-flowcell cartridge. In addition, multi-microwell-array-flowcell cartridges are disclosed. Embodiments of the subject cartridges include a plurality of fluidic lanes, each fluidic lane comprising, an inlet for receiving liquid, a flow cell comprising a microwell array, and an outlet for discharging liquid.

Abstract: A device for agitating and collecting biological liquid samples comprises an agitator of racks of tubes, a sampling apparatus capable of collecting a biological liquid sample in a tube, and a changer capable of gripping a tube on a rack received in the agitator and moving it to the sampling apparatus. The agitator is capable of agitating at least three racks simultaneously, and the device also comprises a scheduler capable of determining destination data for a tube and destination data for the rack which receives this tube, and of determining for each tube a final location based on the destination data of the tube and the destination data of the racks received in the device, which final location designates a rack, received on the agitator and a position on this rack and can be different from the location of the tube when the rack that received it has been introduced into the device, and arranged to control the changer in order to grip a tube, present it to the sampling apparatus and replace it after sampling at the final location.

Abstract: The present invention is directed to sample test cards having an increased sample well capacity for analyzing biological or other test samples. In one embodiment, the sample test cards of the present invention comprise one or more fluid over-flow reservoirs, wherein the over-flow reservoirs are operatively connected to a distribution channel by a fluid over-flow channel. In another embodiment, the sample test cards may comprise a plurality of flow reservoirs operable to trap air thereby reducing and/or preventing well-to-well contamination. The test card of this invention may comprise from 80 to 140 individual sample wells, for example, in a test card sample test cards of the present invention have a generally rectangular shape sample test card having dimensions of from about 90 to about 95 mm in width, from about 55 to about 60 mm in height and from about 4 to about 5 mm in thickness.

Abstract: For enhanced selection of efficient targeted genome manipulating agents, an apparatus includes first and second chip-based biosensors having one or more sensing surfaces configured to detect biomolecular binding interactions between a nucleic acid sample and a targeted genome manipulating agent functionalized to a capture surface within a sensing range of the one or more sensing surfaces. The first chip-based biosensor uses a nucleic acid sample incubated with a blocking agent that blocks on-target binding and the second chip-based biosensor holds a nucleic acid sample that omits the blocking agent. A measurement apparatus measures first and second sets of response signals produced in response to the biomolecular binding interactions occurring between the nucleic acid sample and the targeted genome manipulating agent. An analysis module determines the genome manipulating efficiency parameters of the targeted genome manipulating agent. A system and a method perform the functions of the apparatus.
