摘要: Filamentous fungal host cells expressing hypoxia responsive morphology factor A (hrmA) and biofilm architecture factor (baf) proteins are provided. Methods of producing filamentous fungal host cells expressing hrmA and baf proteins are also provided. In one aspect, the disclosure provides a filamentous fungal host cell, comprising a nucleotide sequence encoding an Aspergillus fumigatus hypoxia responsive morphology factor A (hrmA) protein, or a homolog or ortholog thereof.

摘要: The present invention relates to a novel regulatory T cell protein. This protein, designated PD-L3 OR VISTA resembles members of the PD-L1 family, identified a novel and structurally-distinct, Ig-superfamily inhibitory ligand, whose extracellular domain bears homology to the B7 family ligand PD-L1. This molecule is designated as PD-L3 OR VISTA or V-domain Immunoglobulin Suppressor of T cell Activation (VISTA). Expression of VISTA is primarily within the hematopoietic compartment and is highly regulated on myeloid APCs and T cells. Therapeutic intervention of the VISTA inhibitory pathway represents a novel approach to modulate T cell-mediated immunity for the treatment of a wide variety of cancers, e.g., ovarian, bladder cancer and melanomas. Also, VISTA proteins, especially multimeric VISTA proteins and antibodies may be used to suppress T cell immunity in autoimmune disease, allergy, infection and inflammatory conditions, e.g. multiple sclerosis and arthritic conditions such as RA.

摘要: A haptic stimulator includes a multilayer sheet with a piezoelectric or electroactive polymer layer adapted to mechanically deform upon application of voltage, the multilayer sheet secured to a substrate, and a source of electrical stimulation coupled to drive electrodes on the polymer layer with an AC signal to vibrate the polymer layer. In particular embodiments, the polymer contains polyvinylidene fluoride, and electrodes are patterned to control local electric fields. Another haptic stimulator has first and second electrodes with an air gap and an insulating sheet between first and second electrodes, with an AC voltage driver connecting to the electrodes. In a method of providing haptic stimulation to skin an alternating current supply drives first and second electrodes, the electrodes disposed upon either a piezoelectric or electroactive polymer sheet, vibrating the polymer layer by driving the electrodes; and coupling vibrations of the polymer layer to the sensate skin.

摘要: A method of generating corrected fluorescence data of concentrations of a targeted fluorophore in tissue of a subject includes administering first and second fluorescent contrast agents to the subject, the first contrast agent targeted to tissue of interest, the second agent untargeted. The tissue is illuminated with light of a first stimulus wavelength and first data is acquired at an appropriate emissions wavelength; the tissue is illuminated at a second stimulus wavelength and second data is acquired at a second emissions wavelength associated with the second agent, the first and second emissions wavelength differ. Difference data is generated by subtracting the second data from the first data. A system provides for stimulus and capture at multiple wavelengths, with image storage memory and subtraction code, to perform the method. Corrected data may form an fluorescence image, or is used to generate fluorescence tomographic images.

摘要: The invention is directed to modified T cells, methods of making and using isolated, modified T cells, and methods of using these isolated, modified T cells to address diseases and disorders. In one embodiment, this invention broadly relates to TCR-deficient T cells, isolated populations thereof, and compositions comprising the same. In another embodiment of the invention, these TCR-deficient T cells are designed to express a functional non-TCR receptor. The invention also pertains to methods of making said TCR-deficient T cells, and methods of reducing or ameliorating, or preventing or treating, diseases and disorders using said TCR-deficient T cells, populations thereof, or compositions comprising the same.

摘要: A system and method for depth-resolved imaging of fluorophore concentrations in tissue uses a pulsed light source stimulus wavelength to illuminate the tissue; and a time-gated electronic camera such as a single-photon avalanche detector camera to observe the tissue in multiple time windows after start of each light pulse. A filter device is between the tissue and the electronic camera with fluorescent imaging and stimulus wavelength settings. an image processor receives reflectance images and fluorescent emissions images from the time-gated camera and processes these images into depth and quantity resolved images of fluorophore concentrations in the tissue. Then image processor derives a fluorescence lifetime signal from the received temporal fluorescence signals and derives from these fluorescence lifetime signals biochemical property images of the tissue.

摘要: A method of making a plasmon-resonance biosensor includes conjugating precious metal nanorods with form factor at least 1.5 with a biological probe material. Microfluidic chambers of volume under 0.025 microliter are formed over a substrate, and an aqueous suspension of the conjugated nanorods is injected into the chambers. The nanorods are organized in rows and aligned in long dimension along the rows. The biosensor is configured to be read by obtaining an optical absorption spectrum upon exposure to the analyte. The biosensor includes precious metal nanorods organized in rows with long dimension approximately parallel to the rows. The nanorods are conjugated with biological probes capable of binding to an analyte, the probes may be an aptamer, an antibody, a protein-nucleic acid (PNA), a complimentary DNA, or an enzyme having a binding site.

摘要: A surgical guidance system has two cameras to provide stereo image stream of a surgical field; and a stereo viewer. The system has a 3D surface extraction module that generates a first 3D model of the surgical field from the stereo image streams; a registration module for co-registering annotating data with the first 3D model; and a stereo image enhancer for graphically overlaying at least part of the annotating data onto the stereo image stream to form an enhanced stereo image stream for display, where the enhanced stereo stream enhances a surgeon’s perception of the surgical field. The registration module has an alignment refiner to adjust registration of the annotating data with the 3D model based upon matching of features within the 3D model and features within the annotating data; and in an embodiment, a deformation modeler to deform the annotating data based upon a determined tissue deformation.

摘要: Disclosed is a method for increasing susceptibility of cancer cells to ionizing radiation by delivering to the cells a radiosensitizing agent that has one of the following properties: (a) it perturbs the process of chromosome segregation thereby increasing chromosome missegregation; or (b) it is an inhibitor of an agent that promotes faithful chromosome segregation induces numeric chromosome instability in said cells and this instability is induced substantially simultaneously with or closely prior to or closely after irradiating the cells. Examples of such radiosensitizing agent include inhibitors of one or more of the following: Kif2b, MCAK, MPS1, Eg5/Kinesin-5 5, Polo-like kinase 4, MCAK, Bub1 and Hec1. Such agents specifically target proteins involved in maintaining or promoting faithful chromosome segregation.
