A method for preparing aminoacyl- and misaminoacyl-tRNAs utilizing a specific aminoacyl-tRNA hydrolase is provided. According to the present method, aminoacyl-tRNAs consisting of a tRNA and a corresponding amino acid, or misaminoacyl-tRNAs consisting of a tRNA and a non-corresponding amino acid or an amino acid derivative, can be prepared. In a specific embodiment, L-tyrosyl-tRNA.sup.Tyr is prepared using aminoacyl-tRNA hydrolase from Fusarium culmorum or Artemia salina. In another embodiment a misacylated tRNA, L-phenylalanyl-tRNA.sup.Tyr, is prepared using the aminoacyl-tRNA hydrolase.

The present invention relates to a novel recombinant antibody having a binding activity to 2,3,4,7,8-pentachlorodibenzofuran (2,3,4,7,8-PeCDF), a gene encoding its amino acid sequence, a vector incorporating said gene, a transformant transformed with said vector, a process for preparing said recombinant antibody, and methods for immunologically capturing and determining 2,3,4,7,8-PeCDF using said recombinant antibody. Using the recombinant antibody according to the present invention, it is possible to capture and determine dioxins, in particular, 2,3,4,7,8-PeCDF by an immunological technique in a rapid, convenient, and highly sensitive manner.

The object of present invention is to provide an immunoassay for dioxins which can rapidly and simply afford measured values having a good correlation with analytical values of dioxins by the official method (GC/MS method). The above object is achieved by using the monoclonal antibody of the present invention having not only a reactivity with the indicator isomer among 17 kinds of PCDDs and PCDFs each having a predetermined WHO-TEF value, but also a high cross-reactivity with several kinds of dioxin isomers having five or six chlorine atoms which contribute largely to a TEQ value, and also having a stable reactivity with the antigens in a measuring solvent.
